Objective:To observe the effect of phytostetol ester in cognitive function of high cholesterol diet aged rats,and study its main possible mechanisms,aim to provide experimental basis of application development of phytosterol esters in preventing cognitive impairment.Methods:After Morris water maze test screening,12 months age female SD rats were randomly divided into three groups:control group,high cholesterol group,high cholesterol+PSE group.After 24 weeks intervention,conduct water maze test,evaluate the ability of learning and memory by the escape latency.At the same time determined the serum lipid content at 4 weeks and 24 weeks.Executed after the experimental,observe the pathology change of hippocampal,the cell morphology change in hippocampal CA1 area and number of astrocytes.Detect content of super oxide dismutase(SOD),catalase(CAT),malondialdehyde(MDA),glutathione(GSH);Determination the levels of brain acetylcholine(Ach),cholinesterase(ACh E)and acetylcholine transaminase(Ch AT).Results:After intervention,Control group significantly shorten the incubation period compared with before intervention(P < 0.05),and high cholesterol group significantly prolong the incubation period(P < 0.05),compared with control group,other groups after intervention the escape latency of rats were significantly extended(P < 0.05),but high cholesterol + PSE group incubation period shorter than high cholesterol group of rats.High cholesterol group rats tissue inflammatory cell infiltration,and mild steatosis,high cholesterol group brain hippocampus CA1 area astrocytes(AS)number is more can be seen from the the GFAP dyeing.MDA level of high cholesterol group and high cholesterol + PSE group rats had significant difference(P < 0.05),CAT and GSH levels are lower compared with the control group in different degree.High cholesterol and high cholesterol + PSE group rats Ach and ChAT level had significant difference(P < 0.05),while the Ach E levels higher than the control group with high cholesterol + PSE group.Conclusion:High cholesterol diet can increased blood lipid and damage learning and memory ability of elderly rats,diet with PSE can reduce serum lipid levels,and ease of cognitive impairment in rats and its possible mechanism of action is increase antioxidant capacity and inhibition high cholesterol diet induced antagonism of cholinergic nerve secretory.
