Aim: Hepatocellular carcinoma(HCC)is one of the most common adult malignancy type,even though considerable achievements has been made about treatment in recent years,mortality remains high due to recurrence or liver metastasis.Therefore,it need to strengthen the research of molecular mechanism of invasion and metastasis in HCC,find new targets to prevent and treat HCC,reduce mortality and improve the prognosis.LOX-like protein 2(LOXL2)is one of the most comprehensive studied tumor regulator.Recently,more and more evidence suggested that LOXL2 was involved in invasion and metastasis of various tumor,such as colon cancer,esophageal cancer,breast cancer,and was associated with poor prognosis.But the research about LOXL2 and liver cancer is still small.The present study was to investigate the expression LOXL2 protein in primary liver cancer clinically,explore its relationship with clinical features and the role in invasion and metastasis.Methods: A total number of 80 patients diagnosed with primary liver cancer were collected from the First Affiliated Hospital of Dalian Medical University(Dalian,China)from January 2010-June 2015.The clinical and pathological diagnosis,histological grade and tumor node metastasis(TNM)classification of liver cancer in line with the diagnostic criteria of Chinese "Diagnosis and Treatment of Primary Liver Cancer Standards" 2011.Patients with cirrhosis were made Child-pugh classification.Through hospital cases query system,we collected general condition of patients(sex,age),clinical information(HBs Ag,AFP,presence or absence of liver cirrhosis,Child-pugh classification),and HCC process-related information(tumor size,tumor number,with or without microscopic tumor thrombus,histological grade,TNM stage),each part grouped respectively.Immunohistochemical method was used to detect the expression of LOXL2 protein in tumor tissue(TT)and adjacent non-tumore tissues(ANT)(from the adjacent cancer tissue ?2cm).Quantitative analysis the expression of LOXL2 protein with IPP software,using SPSS 19.0 statistical software to analysis statistical data,and compare the difference expression of LOXL2 between TT and ANT in corresponding groups.Results: 1.LOXL2 protein major stained in the cytoplasm,and LOXL2 expression was higher in ANT than in TT,the difference had statistical significantce(P< 0.05).2.About general condition of patients,LOXL2 expression of ANT was higher in male group than female group,and LOXL2 expression of TT was lower in male group than female group,both the difference had no statistical significantce(P> 0.05);LOXL2 expression of ANT and TT were higher than in the age of < 50 age group than that aged> 50 years old group,the difference had no statistical significantce(P> 0.05).3.About clinical information,LOXL2 expression of ANT was higher in HBs Ag(+)group than HBs Ag(-)group,LOXL2 expression of TT was lower in HBs Ag(+)group than HBs Ag(-)group,the difference had no statistical significance(P> 0.05);LOXL2 expression of ANT and TT were higher in AFP?20ng/ ml group than AFP <20ng /ml group,the difference had no statistical significance(P> 0.05);LOXL2 expression of ANT was higher in liver cirrhosis group than without cirrhosis group,LOXL2 expression of TT was lower in liver cirrhosis group than without cirrhosis group,the difference had no statistical significance(P> 0.05);LOXL2 expression of ANT was higher in Child-pugh classification A group than B group,LOXL2 expression of TT was lower than B group,the difference had no statistical significance(P> 0.05).4.About HCC process-related information,LOXL2 expression of ANT and TT were higher in tumor number ?2 group than one group,the difference had no statistical significance(P> 0.05);LOXL2 expression of ANT was highest in tumor size group 3,LOXL2 expression of TT was lowest in group 2,and differences among the three groups had no statistical significance(P> 0.05);LOXL2 expression of ANT and TT were higher in vascular tumor thrombus group than without vascular tumor thrombus group,the difference had no statistical significance(P> 0.05);LOXL2 expression of ANTand TT increased gradually with histological grade increased,the difference had statistical significantce(P <0.05);LOXL2 expression of ANT and TT increased gradually with TNM stage increased,the difference had no statistical significantce(P> 0.05).Conclusions: 1.The expression of LOXL2 was higher in ANT than TT in primary liver cancer,it means that LOXL2 is conducive to invasion and metastasis.2.The expression of LOXL2 protein has some certain trend with general condition of patients,clinical information,tumor size,tumor number and vascular tumor thrombus,which need further study.3.LOXL2 expression is associated with the malignant degree of tumor,its expression increased gradually with histological grade and TNM stage increased,its high expression contribute to promoting invasion and metastasis in HCC.
