Objective:The main pathogenesis of Hashimoto's thyroiditis (HT) is autoimmune.Studies have shown that miRNA499 (rs3746444) T/C polymorphism can influence inflammatory reaction pathways both in domestic and overseas foreign, then influenceing the development of autoimmune diseases. Our objective of this study is to investigate whether miRNA499 (rs3746444) T/C polymorphism can as a molecular markers of high-risk population in the diagnosis of Hashimoto's thyroid disease of Chinese Han population in Jiangxi,and to provide a theoretical basis for the diagnosis of Hashimoto's thyroiditis.Methods:In this study,a total of 176 subjects from the jiang xi local han people were enrolled from March 2014 to February 2016,including 96 patients with HT which are select from the first affiliated hospital of nanchang university endocrinology and 80 healthy people come from medical department outpatient service.The experimental group was divided into TPOAb? 600 IU/mL and TPOAb< 600IU/mL two subgroups.The study using direct sequencing and polymerase chain reaction-restriction fragment length polymorphism (PCR-RFLP) methods to comparative miRNA499rs3746444 polymorphisms genotypes in the frequency and distribution between the two groups.Results:1.There are three genotypes CC, CT and TT of miRNA499 gene rs3746444T/C polymorphism in experimental group and control group.but there were no statistically significant differences in genotype between the two groups.TT VS CC+CT gene frequency distribution between the two groups have statistically significant differences(P<0.05).T, C allele frequency distribution between the two groups have statistically statistical significance(P<0.05).2.The gene frequency distribution and allele frequency between TPOAb?600 IU/mL group and TPOAb<600IU/mL group has no statistically statistical significance (P>0.05).Conclusion:1.The miRNA499(rs3746444) polymorphism was related with hashimoto's thyroiditis disease of jiang xi local han people.2.Taking CC and CT genotype or C allele of the miRNA499 rs3746444 polymorphism haveing a higher incidence of hashimoto's thyroiditis, prompt that C allele might be the risk factor of hashimoto's thyroiditis, miRNA499 (rs3746444) polymorphism may as a screening molecular markers of high-risk population of hashimoto thyropathy.
