Objective:To study the expression of PD-L1 and TLR4 in non-small cell lung cancer(NSCLC)?Analyze the correlation between the expression of PD-L1 and TLR4 in NSCLC and discuss the associations between the expression and clinicalpathological indexs and prognosis of the patients. Method:The expression of PD-L1, TLR4 were measured in 60 cases of NSCLC, 20 cases of normal lung tissues adjacent to lung carcinoma by immunohistochemistry stain method. Result:1. Expression of PD-L1,TLR4 in non-small cell lung cancer and the control group: The positive rates of PD-L1,TLR4 protein in NSCLC were 61.7%(37/60) and 51.7%(31/60). The control group were 10%(2/20) and 5%(1/20).The difference has statistically significant(P<0.05).2.The relationship between PD-L1,TLR4 expression and clinical-pathological features: The over-expression of PD-L1 suggested the patient has greater probability of recurrence and metastasis(P>0.05).But the expression of PD-L1 has no significant correlation with NSCLC patient gender, age, pathological type, differentiation degree,1ymph node metastasis, TNM clinical staging(P>0.05).TLR4 expression has close correlation with pathological type and clinical staging(P<0.05).But with no significant correlation with NSCLC patient gender, age, differentiation degree,1ymph node metastasis(P>0.05).The over-expression of TLR4 also suggested the patient has greater probability of recurrence and metastasis( P > 0.05).3. The correlation between the expression of PD-L1 and TLR4 in NSCLC: The correlation analysis showed that the two variables were positively correlated(r=0.335,n=60,P=0.009);4. The relationship between the expression of PD-L1, TLR4 in NSCLC with prognosis of lung cancer: Kaplan-Meier survival indicated PD-L1 negative NSCLC patients had longer overall 5-year survival time compared with PD-L1 positive ones(P=0.025);and the expression of TLR4 had the same outcome(P =0.031). Conclusion:The overexpression of PD-L1 ?TLR4 in NSCLC is related with the greater probability of recurrence and metastasis. TLR4 expression has close correlation with pathological type, TNM clinical stage. The expression of PD-L1 and TLR4 protein in non-small cell lung cancer tissues were positively correlated. Overexpression of them in NSCLC indicated poor prognosis.
