ObjectiveTo explore the effect of Lycium Barbarum Polysaccharide(LBP)on restraining the mouse pancreatic cancer cells LTPA by the polarization of macrophages to type 1 macrophages.MethodsConstruct the subcutaneous LTPA tumor model of the CB-17 SCID mice and then the mice were randomly divided into tumor-bearing model group(10 mice)and LBP treatment group(10 mice).The LBP treatment group was fed the dose of 10mg/kg LBP every day,and the tumor-bearing model group was fed the same dose of normal saline.The groups with the same amount of macrophages Raw264.7 were randomly divided into the control and experimental groups of 10,50,80,100,150 and 200 mg/L concentrations of LBP,and MTT was used to detect the optical density(OD)of Raw264.7 in experimental groups and control group;Use the method of ELISA to detect the levels of the interleukin(IL)-12 and IL-10 that Raw264.7 secretes in both the LBP group which LBP was 100 mg/L and the control group.Use the method of flow cytometry to test the levels of the membrane protein CD16/32 and CD206 in both the LBP group which LBP was 100 mg/L and the control group;After feeding 3 weeks,weight and calculate the volumes of the tumors to test the effect of LBP on the growth of subcutaneous tumor.Detect the effect of LBP on the microscopic changes of the tumor by HE staining and the proliferation of the LTPA by KI-67 staining.ResultsThe dose of 100 mg/L LBP can observably promote the growth of the macrophages(P<0.01),and induce macrophages membrane protein higher expression of CD16/32 and lower expression CD206,higher secretion of IL-12 and lower secretion of IL-10.The weight?volume of the tumor and the expression quantity of KI-67 in the experimental group were lower than those in the control group(P<0.01),and the microscopic necrosis area range of the tumor was larger than that in the control group.ConclusionThe LBP has the effect of restraining LTPA by the polarization of macrophages to M1.
