Objective To investigate the effect of Fangji Huangqi decoction on myocardial fibrosis induced by isoproterenol in rats and its possible mechanism.Methods 63 SD rats are assigned to the control group,model group,captopril group,and Fangji Huangqi decoction treatment group(low,middle and high dose),and the low,middle and the high dose groups were used as standard dose of 1/2,1 and 2 times respectively,and the control group was 8,and the other groups were 11.Except the control group,other 5 groups were back multi-point injection of isoproterenol method(5 mg·kg-1·d-1×7d)to establish the model of myocardial fibrosis,and the control group were injected with saline of the same volume.Intragastric administration were began after 7 days,and it lasted 4 weeks.4 weeks later,hemodynamics changes of heart were tested,the heart weight index and left ventricular weight index were calculated;the morphologic changes of myocardial were observed by Masson staining;the content changes of IL-1?,IL-6,IL-10 and TNF-? were measured by ELISA;the changes of TGF-?1 and P-I?B? in myocardial tissue of rats were tested by Western blot.Results Compared with the model group,the middle and high dose groups of Fangji Huangqi decoction can obviously improve heart function of rats,depress the heart weight index and left ventricular weight index,reduce the collagen volume fraction of myocardium,depress the levels of IL-1?,IL-6 and TNF-?,raise the IL-10 levels,depress the expression of myocardial TGF-?1 and P-I?B?;compared Fangji Huangqi decoction middle dose to high dose,these indexes were not significant difference;the low dose grop only could reduce the levels of IL-1? and TNF-?,on rat heart function,HWI,LVWI,CVF,IL-6,IL-10,TGF-?1 and P-I?B?,there was no significant change.Conclusions Fangji Huangqi decoction can inhibit the myocardial fibrosis induced by isoproterenol in rats,and its mechanism may be related to the down-regulation of the expression of IL-1?,IL-6,TNF-? and P-I?B? in NF-?B signal pathway.
