ObjectiveTo investigate the effects of moxibustion on the learning and memory abilities,A?42,Bcl-2,Bax and IL-1? of AD rats,and explore the effect and the possible mechanism to AD of moxibustion in protecting system.Methods A total of 60,2 months healthy male SD rats were divided randomly into 5 groups with 12 rats in each group: normal group,sham-operated group,Alzheimer's disease model group,drug group and moxibustion group.Rats of the normal group of conventional feed without any treatment,the sham group are given citric acid/sodium citrate buffer by lateral ventricle injection,and the rats of the after three groups are injected with the same vilume of STZ into the lateral ventricle to construct AD model.After the operated,The Morris water maze was used to observe the learning and memory ability for the rat again.As compared with the before,the after have lengthen the average of escape latency and dropped the across platforms,which proved the model was succeed.And then,the two of died rats was excluded,the rest of rats was divided into groups: normal group(n=12),sham-operated group(n=12),Alzheimer's disease model group(n=11),drug group(n=11)and moxibustion group(n=12).Moxibustion group was respectively given at :“Baihui”(GV20),“Mingmen”(GV4),“Changqiang”(GV1)and “Guanyuan”(CV4),Mild moxibustion(once alternate daily,One period of treatment for 10 times,3 times altogether).The drug group are given the donepezil Hydrochloride Tablets of 0.45mg/kg as the same course of moxibustion treatment.The Morris water maze was used to observe the learning and memory ability for the rat.Immunohistochemical was used to detect the expression of A?42 in hippocampus.Western Blot was used to detect the expression of Bcl-2 and Bax in hippocampus.Biochemical methods were used to determine the content of IL-1? in the hippocampus tissue.Results 1.Morris water maze test results: As compared with the sham group,the normal group have no significant different(P>0.05),while the model group have lengthen the average of escape latency and dropped the across platforms(P<0.05);in comparison with the model group,the drug and moxibustion could obviously shorter average escape latency and more crossing times(P<0.05);compared with the drug group,moxibustion group had no statistical differences in the average of escape latency and across the platform(P>0.05).2.Immunohistochemical results: As compared with the sham group,the normal group have no significant different in terms of the expression of A?42(P>0.05),while the model group have increased the expression of A?42 positive cells(P<0.05);in comparison with the model group,the drug and moxibustion could obviously shorter the expression of A?42 positive cells(P<0.05);compared with the drug group,moxibustion group had shorter the the expression of A?42 positive cells(P<0.05).3.Western Blot test results: The expression of apoptosis related proteins,Bcl-2 and Bax,there was no significant difference between Sham group and Normal group(P>0.05);The expression of Bcl-2 in the model group were significantly decreased relative to the sham groups(P<0.05),and the Bax increased marked(P<0.05);The relative expression of Bax in drug and moxibustion group were less than the model group(P<0.05),and the Bcl-2increased(P<0.05);The expression of Bax in moxibustion group were less than the drug group(P<0.05),and the Bcl-2increased(P<0.05).4.Biochemical methods test results: The expression of IL-1? has no significant difference between Sham group and Normal group(P>0.05);As compared with the sham group,the model group has increased the expression of IL-1?(P<0.05);in comparison with the model group,the drug and moxibustion could obviously shorter the expression of IL-1?(P<0.05);compared with the drug group;compared with the drug group,moxibustion group had decreased the expression of IL-1?(P<0.05).Conclusions AD model rats were established by lateral intracerebroventricular injection of STZ,the ability of learning and memory was impaired,and expression of A? 42 ? IL-1? and Bax were increased,while the expression of Bcl-2 were decreased;The treatment by given moxibustion and drug can improve the learning and memory ability of AD model rats,but the moxibustion as more effective as drug;The moxibustion can enhance their learning and memory abilities,may be related to decrease the expression of A?42 ?IL-1? and Bax,white add the levels of Bcl-2 in hippocampus tissue.
