ObjectiveTo identify the role of mi R-145 in endothelial-to-mesenchymal transition(EndMT)of human umbilical vein endothelial cells(HUVECs)which may regulate expression of ADAM17.MethodsHUVECs were cultured and the cell morphology was observed using phase contrast microscope.Mi R-145 which may regulate expression of ADAM17 was predicted using bioinformatics.The expressions of mi R-145 ? ADAM17VE-cadherin and ?-SMA were determined using real-time PCR and western blotting after transfection of mi R-145 mimics.ResultsExpression of VE-cadherin was positive before the induction of TGF-?1and after induction ?-SMA was found by immunofluorescence cytochemistry.Results of real-time PCR and western blotting showed that over-expression of mi R-145 up-regulated expression of VE-cadherin and down-regulated expression of ADAM17 and ?-SMA.ConclusionsMi R-145 may negatively regulate expression of ADAM17 and inhibit EndMT of HUVECs. |
