ObjectiveThis study aims at observing the phosphorylated(p)extracellular signal-regulate kinase(ERK)and cAMP-response element binding protein(CREB)distribution in the hippocampus of mice on embryonic(E)18th day(d),postnatal(P)1th d,3th d,7th d,14 th d,21 th d and 28 th d.Our study will support some basal experimental datas for the study of mechanism of hippocampus development.MethodsKunming white mice were selected in this experiment.Female and male mice were fed in the same case with the ratio of 1 to 2.The day when the vagina plug was found was looked as the embryonic 0 day.The day when the baby mice were born was the postnatal 0 day.The hippocampus of mice on E18 d,P1d,P3 d,P7d,P14 d,P21d and P28 d were obtained to study,6 mice in each group.Immunohistochemistry,Western blot and image analysis methods were used to analyze qualitatively and quantitatively the expression of pERK and pCREB in the development of hippocampus at different stages.ResultsImmunohistochemistry staining revealed that pERK and pCREB were mainly expressed in the nervous cell nucleus of pyramidal layer of CA area and dentate gyrus(DG)granular layer of hippocampus.The pERK and pCREB positive cells arranged sparsely,slight staining,with the decrease of optical density in CA and DG area on E18 d.From E18 d to P7 d,the positive cells increased gradually,with dark staining and the increase of optical density,especially went to the top on P7d(P<0.01).Then the expression of pERK and pCREB and optical density of P14 d to P21 d began to decrease gradually(P<0.01).The positive expression of P28 d was similar to that of P21d(P>0.05).Western blot revealed that the expression of pERK and pCREB increased gradually from E18 d to P7d(P<0.01),then decreased during P14 d to P21d(P<0.01)and stabilized on P28d(P>0.05).ConclusionspERK and pCREB are related to the development of mice hippocampus.pERK and pCREB take part in the cell proliferation in the development of mice hippocampus.
