Expression Of Osteopontin And Integrin ?v?3 In The Retina Of Diabetes Mellitus With High Myopia Guinea Pigs

Objective: This research established the model of diabetes mellitus with form-deprivation myopia guinea pigs,using immunohistochemistry method to observe the expression of osteopontin(OPN)and integrin ?v?3 in the retina.To explore the effect of high myopia on osteopontin(OPN)and integrin ?v?3 levels in diabetes mellitus.For further investigate the mechanism and significance of interaction between high myopia and diabetes mellitus.Methods:Four groups of guinea pig model is established in this experiment.Selectd fifty England healthy one week old guinea pigs randomly divided into 4 groups,excluded congenital eye diseases and ametropia.Chosen eight guinea pigs as the blank control group(group ?)which were injected buffer solution intrapeitoneally and ordinary feeded for 20 weeks.The rest were divided into three groups,every group have twelve guinea pigs.Group ? is form-deprivation myopia guinea pigs by using non-invasive mask cover the right eye for twenty weeks and at the eighth weeek they should be injected buffer solution intrapeitoneally as 280mg/kg.Group ? is diabetes mellitus guinea pigs.After fasting for 24 hours,intraperitoneal injection streptozotocin(STZ)280mg/kg.Group ? is diabetes mellitus with form-deprivation myopia guinea pigs,the method is just combine the front two methods.The refraction diopter should at least >-6.00 D and the blood glucose ? 16.5mmol/L after intraperitoneal injection of STZ for 72 h.If the blood glucose lower than16.5mmol/L,add intraperitoneal injection streptozotocin(STZ)180mg/kg.Three groups were ordinary feeded for twelve weeks after model.Ater the model experiment,selected eight guinea pigs from each group,then sacrificed all the thirty three guinea pigs and retinal tissues were obtained.Given HE stainning and immunohistochemistry to detect the expression of OPN and integrin ?v?3 in retina respectively and observe the order and cells number of the retina.Preliminary discuss the mechanism and significance of interaction between high myopia and diabetes mellitus.Results:1.The average weight of non-diabetes group(group ? and group ?)were significantly higher than diabetic diabetes group(group ? and group ?),the difference have statistical significance(p<0.05).Average glycemic of non-diabetes group(group ? and group ?)were significantly lower than diabetic diabetes group(group ? and group ?),the difference have statistical significance(p<0.05).2.The average refraction diopter of group ? and group ? were significantly lower than diabetic group ? and group ?,and the axis oculi is just on the contrary,also the difference have statistical significance(p<0.05).3.OPN and integrin ?v?3 in retina is expressed by the ganglion cell layer,The average expression level of diabetic group(group ? and group ?)were significantly higher than non-diabetic group(group ? and group ?),the difference have statistical significance(p<0.05).Including,group ? was significantly higher than diabetic group ?,the difference have statistical significance(p<0.05),too.4.Observe the retina tissue by HE staining,The thickness,shape,arrangement,and the number of cells were normal in group ?.The retina thickness of group ? is lower than group ?,and the order is mess,the gap between each cells are expand.Group ? and ? have a lower cells number than namal,and the retina ganglion cell layer appeared degeneration and necrosis.Part of the retina have fracture.Conclusion:1.The expression level of OPN and integrin ?v?3 in diabetic group(group ? and group ?)were significantly higher than non-diabetic group(group ? and group ?).2.Combine non-invasive mask with intraperitoneal injection streptozotocin(STZ)280mg/kg can establish the model of diabetes mellitus with form-deprivation myopia guinea pigs succesfully.3.High myopia is a protective factor of diabetic retinopathy,which can delay the duration of diabetes mellitus.
