| Objective: This study was to explore the effect of Ang and Tie 2 in the treatment for infantile hemangioma in nude mice model with propranolol,Clear purpose is to further explore the mechanism of propranolol in the treatment of hemangioma,and lay the foundation for clinical work.Methods:1 Hemangioma tissue transplantation is used to the proliferation of vascular surgical resection of tumor tissue were transplanted into the back of 50 female nude,build hemangioma nude mice;all experimental mice were randomly divided into two groups,propranolol treatment group and control group,observed changes in the growth of transplanted tumors in each group;2 Mice were sacrificed before treatment and after treatment 3d,6d,9d,14 d,measuring tumor size,pathology make HE staining,immunohistochemist-ry and western blot were used to detect the expression of VEGF,Ang1,Ang2 and Tie2 protein in tumor tissue.Results:1 The tumor volume and HE staining: tumor volume decreases in the propranolol group compared with that before treatment(P <0.05),and a timeand dose-dependent manner,HE staining and found that the number of hemangiomas was significantly reduce,and there are plenty of fat and fibrous tissue.2 Immunohistochemical results: VEGF,Ang2,Tie2 brownish yellow particles in the control group,the increase in the number and expression level increased,while the expression of Ang1 reduction,to reduce the number.Expression of VEGF,Ang2,Tie2 reduction in the propranolol group,while the expression of Ang1 increases.Differentially expressed VEGF,Ang1,Ang2,Tie2 significantly before and after propranolol(P <0.05).3 western blot results: VEGF,Ang2,Tie2 protein expression in the control group increased gradually after drug intervention,and the expression of VEGF,Ang2,Tie2 protein was significantly decreased in the propranolol group(P <0.05);the expression of Ang1 in the control group decreased after the intervention with drugs,while Ang1 expression was significantly increased in the propranolol group(P <0.05).while the expression of Ang1 increases.Differentially expressed VEGF,Ang1,Ang2,Tie2 significantly before and after propranolol(P <0.05).Conclusions: Propranolol inhibition of tumor growth in nude mice blood vessels by promoting the expression of Ang1 and inhibit the expression of VEGF,Ang2,Tie2. |
