Objective: The clinicopathological records of patients with diagnosis of primary salivary gland acinic cell carcinoma(AciCC)were retrospectively reviewed.Detect the expression of Mucin 1(MUC1)in salivary gland acinic cell carcinoma and investigate the relationship between the expression of MUC1 in salivary gland acinic cell carcinoma and its clinicopathological features as well as disease prognosis,in order to provide some new data and recommendations for early diagnosis,clinical treatment and prognostic evaluation of AciCC.Methods:1 The clinical and complete follow-up datas of patients with salivary gland tumor were collected from the department of oral and maxillofacial of the Fourth and the Second Hospital of Hebei Medical University as well as Xingtai People's Hospital from January 2007 to June 2015.Elect and retrospect the clinical datas of the gender,age,location,pathological variant,local recurrence,lymphatic metastasis or distant metastasis and pathological classification of primary salivary glandacinic cell carcinoma.All of the above statistical methods were performed by Microsoft Excel 2007 Software.2 Select the normal salivary gland(NSG)tissue as the control group according to the pathogenic site of Aci CC,and primary salivary gland AciCC tissue is taken as the experimental group.The rate of MUC1 positive cells in the experimental group and the control group is detected by MaxVision immunohistochemistry.Use SPSS 17.0 statistical software for statistical processing,with mean ± standard deviation((?)±s)noted,use independent sample T test to make comparison between two groups,use analysis of variance when paired comparison are required,and use SNK-q test to have comparisons,and P < 0.05 indicates difference is statistically significant.Results:1 A total of 1338 patients with salivary gland tumor are collected,with 52 cases of primary salivary gland AciCC accounting for 4% of the salivary gland tumors and 13% of the malignant gland tumors.Among 52 cases,25 cases for male and 27 cases for female with the male/female ratio 1:1.08 and the average age 48 years old.The highest incidence rate occurs at 51 to 60 years age group,accounting for 48.08%(25/52),and parotid gland appears to be the most frequent sites,accounting for 67.31%(35/52),followed by palate accounting for 11.54%(6/52),submandibular gland site ranks third,accounting for 7.69%(4/52),other small salivary gland area accounts for 13.46%;there are 27 cases of solid type,16 cases of micro capsule type,8 cases of papillary cystic type and 1 case of follicular type of AciCC tissues;the cervical lymphatic metastasis rate is 11.54%(6/52);there are 51 cases followed-up and 1 case missed,with the recurrence rate of 13.46%(7/52)and the distant metastasis rate of 3.85%(2/52).2 MUC1 is only expressed in lumen catheter glandular of the NSG tissues,distributed on the membrane top of the epithelium cells.All AciCC tissues show the expression of MUC1 which is distributed in cytomembrane and cytoplasm of tumor cells as well as in some nucleus with distributed polarity.3 The rate of positive cells of MUC1 in AciCC tissues is(86.4±2.8)%,compared with(9.1 ±0.6)% with NSG tissues(P < 0.05),with statistical significance.4 The rate of positive cells MUC1 in solid type,micro capsule and papillary cystic of AciCC tissues is respectively(92.6±3.1)%,(80.5±2.6)% and(78.2±1.9)%,the comparison among different tissue types of salivary glands AciCC shows P < 0.05.5 The rate of positive cells of MUC1 for female[(90.2 ± 1.8)%] is higher than that for male [(82.3± 2.1)%],the patients of 41 ~ 60 age group [(91.6±3.8)%] is higher than other age groups [0 ~ 20 age group is(85.0±1.8)%,21 ~ 40 age group is(87.5 ±2.3)%,61 ~ 80 age group is(88.3 ±1.7)%],the rate in submandibular gland AciCC[(93.7 ±2.4)% ]is higher than that in other parts [parotid gland group(86.8±2.8)%,palate group(81.3±3.0)%,and other small salivary gland group(83.2±1.4)%],the rate in lymphatic metastasis groups [(88.7 ±1.1)%] is higher than that in those without [(86.1 ±1.8)%],the rate in recurrence or distant metastasis groups [(93.1± 2.6)%] is higher than that in those without [(85.0± 2.9)%],the comparison between groups shows P < 0.05,with statistical significance.Conclusions:1The expression of MUC1 in the salivary gland AciCC tissue is significantly different with the NSG,and there is a significant difference between the rate of positive cells and NSG tissues,showing that MUC1 is related to the occurrence and development of AciCC.2The expression level of MUC1 in AciCC is related to gender,age,tissue type,pathogenic site,lymphatic metastasis,local recurrence and metastasis,showing that MUC1 may be a new tumor marker of salivary gland AciCC,so MUC1 probably has important clinical value on clinical diagnosis,treatment and prognostic judgment of salivary gland AciCC.
