Objective: The harm problem of bone marrow failure with iron overload has caused more and more attention,and the researches related have become a hot spot,there is still lack of index and therapeutic strategy of the aplastic anemia(AA)in the consensus about the diagnosis and treatment of the iron overload in China.This research aims at probing the realations of the Hepcidin and SF in changes and transfusion and the new treatment of iron overload disorders in children with anemia by detecting the levels of the Hepc,IL-6,EPO,SFand TRF and comparing the realationships of Hepcidin,SF,IL-6,EPO and TRF.Methods:1 Cases and Groupingrandomly select 30 aplastic anemia children as case group who have been hospitalized in the second hospital of hebei medical university's pediatrics in blood and oncology,17 children whose blood transfusion no less than 8U for group A,the other 13 whose blood transfusion no more than 8U for group B,meanwhile select 30 healthy children who has physical examination in the hospital as comparison group according to their gender,ages and other factors matched,and the 30 children in comparison group do not have diseases in blood,circulatory,endocrine,nervous system and mental disorders,and except for cancer and infectious diseases.2 Specimen collection,specimen handling and detection of related dataAll of the subjects should be collected serum samples 5ml on an empty stomach in the morning,all the specimens were collected in coagulant in the tube,then make a speed centrifugal for 10 minutes with ordinary centrifuge at 3000 r / min,collected serum samples,then send the specimens of SF and TRF to hospital immediately to be detected,use electro-chemiluminencesce immumoassay technology for SF detection and rate scattering turbidimetry for TRF detection.The remaining samples are stored in the refrigerator at the temperature-80 ?,serum Hepcidin,IL-6 and EPO levels which are be tested will be measured by ELISA.3 Statistical methodThe data measured is statistically analysed by adopting SPSS21.0 software,all the data of case group and comparison group are measurement data,all data not conforms to normal distribution are showed by Median,test results P<0.05 for the difference statistically significance,P<0.01 can be significant difference.P>0.05 with no difference is not statistically significant.Results: 1 The Hepcidin level(82.7805 ug/L)of case group was significantly higher than the comparison group(10.0175 ug/L),the difference between the two groups was statistically significant(P<0.01),the EPO level of case group(5.9445 ng/L)was significantly higher than the comparison group(2.1070 ng/L),the difference between the two groups was statistically significant(P<0.01),the IL-6 level of case group(13.4795 U/L)was significantly higher than the comparison group(6.3940 U/L),the two groups was statistically significant(P<0.01);2 There is a positive correlation between Hepcidin and SF(The correlation coefficientof Spearman is 0.974 P<0.01),there is a positive correlation between Hepcidin and IL-6(The correlation coefficientof Spearman is 0.637 P<0.01),there is Negative correlation coefficient between Hepcidin and EPO(The correlation coefficientof Spearman is 0.875 P<0.01),there is no correlation between Hepcand TRF(P> 0.05);3 The Hepcidin level of case group whose blood transfusion no less than 8U(98.979 ug/L)is significantly higher than the group whose blood transfusion no more than 8U(43.487 ug/L),the SF level of case group whose blood transfusion no less than 8U(656.90 ug/L)is significantly higher than the group whose blood transfusion no more than 8U(134.50 ug/L),the difference between the two groups was statistically significant(P<0.05).Conclusions:1 The levels of Hepcidin,IL-6 and EPO of the aplastic anemia children are higher than the comparison group,the level of Hepcidin,IL-6 and EPO can be used as an indicator to detect aplastic anemia.2 The level of Hepcidin,SF and IL-6 of the aplastic anemia children has a positive correlation,and has a negative correlation with EPO level.3 The Hepcidin and SF level of the aplastic anemia children whose blood transfusion no less than 8U is significantly higher than the Hepcidin level whose blood transfusion no more than 8U,which explains that Hepcidin and SF will rise with the increase of blood transfusions,when Hepcidin increase the iron absorption can be reduced,thereby reducing the incidence of hemochromatosis,this may be one of the treatments of iron overload.
