Colitis associated with colorectal cancer(CAC,colitis associated colorectal cancer)is a chronic intestinal inflammation associated malignant transformation.The reason for its occurrence and development has not been studied well.Generally,it is accepted that environmental factors cause the sustained activation of colonic immune system.Then,the maintaining intestinal inflammatory microenvironment result in the repeated process of colon tissue damage and repair,finally lead to the malignant transformation of colonic epithelial cells.As a common serious threat to human life and health,the pathogenesis study of CAC has become a hot topic in the medical field.C/EBP-?(CCAAT enhancer binding protein,CCAAT/enhancer binding protein-?)is a multi-directional transcription factor,which participates in both normal physiological metabolism and the disease process,such as tumors and inflammation-related diseases.However,the biological function of C/EBP-? in CAC is not yet clear.To investigate the biological function of C/EBP-? in CAC,we use C/EBP-? gene knock-out mouse to construct mouse CAC model.Both the number of and the size of tumors in C/EBP-? gene knock-out mouse are reduced.By molecular and cell biology techniques,we proved that C/EBP-? mediated pathway(C/EBP-?-miR-31—IRASA1?CAC)played an important role in the development of CAC.The study will not only provide new theoretical evidences for the malignant transformation basis during colitis-associated malignant transformation,but also provide novel targets and therapeutic clues for the clinical treatment of uncontrolled colitis-associated malignant transformation.
