A Study About Traditional Chinese Medicine On Anti-aging And Diabetic Vascular Complication Based On Calorie Restriction

High energy intake can mediate the metabolic syndrome(such as diabetes vascular complications and accelerating aging)occurs.Diabetes is "Xiaoke" in traditional Chinese medicine.Qi-yin deficiency is essence of diabetic vascular complication,while qi stagnation and blood stasis are surface of it.Astragalus(Huangqi,HQ)replenish qi,Salvia miltiorrhiza and Panax pseudoginseng(Danshen and Sanqi,DQ)activitive blood.So we use Astragalus,Salvia miltiorrhiza and Panax pseudoginseng recipe cure the qi stagnation and blood stasis of diabetic vascular complication.In drosopHila,nematodes,mice and other model organisms,high energy intake accelerated the aging of them.However some medicine such as resveratrol and AMPK agonists could partially reverse this pHenomenon.Polygonum multiflorum thunb was considered to have the effect of prolonging life,but its specific effectiveness,material foundation an d molecular mechanism were still not known clearly.Tetrahydroxystilbene glucoside(TSG),an extract of Polygonum multiflorum thumb,had a similar structure to resveratrol.So it was speculated that the medical effectiveness of Polygonum multiflorum thumb was partly based on TSG.It could reverse the fat-mediated accelerated aging by regulating energy metabolism.Obeject:We explore the efficacy of QSYQ on diabetic foot and 2,3,5,4-tetrahydroxyl dipHenylethylene-2-o-glucoside on the aging which was mediated by fat food.Then we want to make a thorough inquiry mechanism of them.Method:High-fat diet-fed unite streptozotocin was used to induce C57 mice type 2 diabetes unilateral hind limb ischemic model.QSYQ,HQ and DQ are gavaged.ACCU-CHEK was used to detect the fasting blood glucose oral glucose tolerance test(OTGG).Laser Doppler Flowmeter was used to test blood flow and its mechanism were explored.After feeding the 12-month-old mice with high-fat food,the weigh of them were measured,Walking patterns analysis experiment,rotarod endurance experiment and bone density was tested periodical.Then the mice were killed to make H&E staining of heart,liver and kidney.Result:Conclusion is that QSYQ improve the blood flow recovered observably about 100%,and its mechanism may be activating EPCs(about 100%)and up regulate angiogenesis-and energy metabolism-relation gene partly.QSYQ and its "Blood-activating",but not"Qi-benefiting" components,restored membrane potential of endothelial cells.In a period of time,TSG could not only reverse the accelerated aging mediated by high-fat food and the decline of endurance,but also the accelerate reduction of bone density and the increase of myocarditis.But it should not be used for a long time because its toxicity.
