Objectives To investigate the influence of the extracts from Penthorum chinese Pursh on pathological changes and therapeutic benefits in cholestasis liver fibrosis rats through TGF-?/Smad signal pathway.Methods: Cholestasis liver fibrosis model was set upby common bile duct ligation of both ends and then a middle cut-off in male SD rats.Total 40 male SD rats with successful modeling were randomly divided into four groups: common bile duct ligation group,high and low dose(4g/kg-1·d-1,2g/kg-1·d-1)of the extracts from Penthorum chinese Pursh groupgroup,ursodeoxycholic acid group?30mg/kg?with 10 rats in each group.Another 10 male SD rats were served as control group randomly.Medicines were all taken by gavage,normal and common bile duct ligation group rats were given equal dose of normal saline.The rats were anaesthetized at the end of 2nd and 4th week after operation to take the blood from heart for detecting not only the serum level of?hyaluronic acid,HA?,?laminin,LN?,?collagen type ?,C??,but also liver function indexes?ALT?AST?ALB?DBIL?TBIL?,meanwhile,partial hepatic tissues of all 5 groups rats were taken for biopsy.If the nude mice died in the experimental process,An autopsy should be done in time.All the hepatic tissues were fixed in 10% neutral formalin,embedded in paraffin and stained with HE to observe the pathological changes of hepatic tissue and the degree of hepatic fibrosis,besides the dyeing parts and staining intensity of TGF-?1?transforming growth factor ?1,TGF-?1?,Smad3,Smad7 and ?-SMA were determined by immunohistochemistry Envision method.Results:?1?Serum Level of HA,LN,pc?,c?among ursodeoxycholic acid group and both high and low dose of the extracts from Penthorum chinese Pursh group were all inferior to those of common bile duct ligation group on 14 th and 28 thday in SD rats,?p<0.05?.?2?Hepatic function indexes?ALT?AST?ALB?DBIL?TBIL?among ursodeoxycholic acid group and both high and low dose of the extracts from Penthorum chinese Pursh group were all inferior to those of common bile duct ligation group on 14 th and 28 thday in SD rats?p<0.05?.?3?The expression levels of TGF-?1,Smad3 and ?-SMA among ursodeoxycholic acid group and both high and low dose of the extracts from Penthorum chinese Pursh group were all inferior to those of common bile duct ligation group on 14 th and 28 thday in SD rats?p<0.05?.?4?Serum Level of HA,LN,pc?,c and ?hepatic function indexes?ALT?AST?ALB?DBIL?TBIL?among ursodeoxycholic acid group and both high and low dose of the extracts from Penthorum chinese Pursh group were not statistically significant at the same time?p>0.05?.?5?Degrees of liver fibrosis among ursodeoxycholic acid group and both high and low dose of the extracts from Penthorum chinese Pursh group were all inferior to those of common bile duct ligation group on 28 thday in SD rats?p<0.05?,with no statistical differences among ursodeoxycholic acid group and both high and low dose of the extracts from Penthorum chinese Pursh group?p>0.05?.Conclusion:?1?The extracts from Penthorum chinese Pursh can slow the progression of cholestasis-induced liver fibrosis in SD rats.?2?The anti-hepatic fibrosis mechanisms of the extracts from Penthorum chinese Pursh may be related to its inhibiting TGF-?1/smad signal transduction pathways and reducing the production of TGF-?1?Smad3??-SMA,etc which were crucial to liver fibrosis.
