this article plans to choose the human acute myeloid leukemia cell line which carries FLT3-ITD(MV4-11)as target,observes the influence on MV4-11 cell cycle and proliferation of rapamine and explore the functions of PI3K/AKT/mTOR signal way,and downstream protein(4E-BP1 and p70S6K)in the process.Methods : According to the different concentrations of rapamine,MV4-11 cells were divided into control group and experimental group,each group was cultured for 24 hours.Then we tested the proliferative activity by CCK-8.Cell distribution in the cell cycle were determined using flow cytometry.western-blot and reverse transcription-PCR were used to detect the mTOR,4E-BP1 and p70S6 K mRNA expression,protein and phosphorylated protein distribution in each group.Results: When its concentration was10nmol/L-200nmol/L,rapamine showed significant growth inhibition effect for the human leukemia cells which carrise FLT3-ITD,IC50 was87.2nmol/L.Meanwhile,the proportion of G0/G1 cells increased significantly(P?0.05).The experimental group under the influence of rapamine(10nmol/L-200nmol/L)showed a lower expression level of p70S6K?4E-BP1 mRNA(0.79±0.15,0.61±0.08,0.42±0.09,0.28±0.06,0.12±0.06 and1.66±0.13,1.23±0.11,0.95±0.09,0.65±0.12,0.35±0.09),.as compared to the control groups(P<0.05).The influence of protein and phosphorylated protein had the similar result.mTOR and p-mTOR protein expression quantity incontrol group was higher than its experimental groups,when the concentration was 50nmol/L-200nmol/L(P<0.05).
