Objective: to detect the expression levels of RUNX3 and STAT3 by immunohistochemistry envision method. To study the mechanism of RUNX3 and STAT3 in the development and growth of gastric cancer and its effect on invasion, metastasis and prognosis of gastric cancer. To analyse the relevance between RUNX3 and STAT3 and clinicopathological characteristics of gastric cancer patients(including age, tumor size, transfer situation etc.). To promote early diagnosis and treatment of gastric cancer clinically. Methods : 120 cases of experimental samples were taken from archive paraffin blocks of pathology dapartment. Including 24 cases of normal gastric mucosa tissue(all the normal gastric mucosa tissues were taken away from the tumor cutting margin of 5cm), 20 cases of atypical hyperplasia gastric tissue( including 8 cases of mild atypical hyperplasia and 12 cases of severe atypical hyperplasia), 76 cases of gastric cancer tissues(including 39 cases of poorly differentiation, 37 cases of medium and high differentiation), immunohistochemical envision method was used to detect the expression of RUNX3 and STAT3 protein in each group. SPSS(19.0 version) statistical software was used for statistical analysis, the positive rate was compared using chi-square test analysis; Spearman correlation analysis method was used for relevance analysis of RUNX3 and STAT3 in gastric cancer; P < 0.05 was thought that difference was statistically significant. Results:(1) In 24 cases of normal gastric mucosa tissues RUNX3 positive expression was found in 22 cases, the positive rate was 91.67%, in 20 cases of atypical hyperplasia tissues RUNX3 positive expression was found in 13 cases, the positive rate was 65%, in 76 cases of gastric carcinoma tissues RUNX3 positive expression was found in 24 cases, the positive expression rate was 31.58%. From the above data it can be seen that the differences of RUNX3 positive rate between the gastric cancer tissues and atypical hyperplasia tissues and normal gastric mucosa tissues were statistically significant(x2=21.720, p<0.05; x2=7.204, p<0.05); the difference of RUNX3 positive rate between atypical hyperplasia tissues and normal gastric mucosa tissues was statistically significant(x2=20.187, p<0.05); in 76 cases of gastric carcinoma tissues STAT3 positive expression was found in 55 cases, the positive rate was 72.37%, in 20 cases of atypical hyperplasia tissues STAT3 positive expression was found in 9 cases, the positive expression rate was 45%, 24 cases of normal gastric mucosa tissues STAT3 positive expression was found in 3 cases, the positive expression rate was 12.5%. The differences of STAT3 positive rate between gastric cancer tissues and atypical hyperplasia tissues and normal gastric mucosa tissues were statistically significant(x2=21.598, P < 0.05; x2=6.241, P < 0.05); The difference of STAT3 positive rate between atypical hyperplasia tissues and normal gastric mucosa tissues was statistically significant(x2=16.301, P < 0.05);(2) To analyse the relationships between RUNX3 and gastric cancer clinical pathological factors : RUNX3 positive expression rate was higher in the highly differentiated group, the positive expression rate was low in the groups of the deeper invasion, lymph node metastasis and highly TNM stage, it indicated that the RUNX3 expression was significantly related with gastric cancer(P<0.05), in addition, there was no relationship between the RUNX3 expression and the age?gender and tumor size of gastric cancer patients(p>0.05); To analyse the relationships between STAT3 and gastric cancer clinical pathological factors : STAT3 positive expression rate was low in the highly differentiated group, the positive expression rates were higher in the groups of deeper invasion, near or distant lymph node metastasis and highly TNM stage, it indicated that the STAT3 expression was significantly related with gastric cancer(P<0.05), in addition, there was no significant correlation between the STAT3 expression and the age?gender and tumor size of gastric cancer patients(p>0.05);(3)The was a negatively correlation between the expression of RUNX3 and STAT3 in gastric cancer tissues by statistical analysis(R; =-0.662, P<0.05). Conclusion:1.From normal gastric mucosa to atypical hyperplasia and gastric carcinoma, RUNX3 positive expression rate shows a gradually decreasing trend, in gastric carcinoma tissues RUNX3 positive expression rate is inversely proportional to malignant degree, the positive expression rates are lower in the groups of poorly differentiation? deeper invasion, lymph node metastasis and highly TNM stage, it indicates that RUNX3 may be helpful to early diagnosis and prognosis evaluation of gastric cancer patients. 2.From normal gastric mucosa to atypical hyperplasia and gastric cancer tissue, STAT3 positive expression rate shows a gradually increasing trend, in gastric carcinoma tissues STAT3 positive expression rate is directly proportional to malignant degree, the positive expression rates are higher in the groups of poorly differentiation?deeper invasion, lymph node metastasis and highly TNM stage, it indicates that STAT3 may be closely related with growth and development of gastric cancer, the detection of STAT3 may be served as a reference index for evaluating the prognosis and survival time of gastric cancer. 3.The expression of RUNX3 and STAT3 in gastric carcinoma is negatively correlated, and the combined detection of two indexes may be helpful to the early diagnosis of gastric cancer and it may play a guiding role in the treatment and prognosis evaluation of gastric cancer.
