| bjective:Inflammatory bowel diseases including Crohn’s disease and Ulcerative colitis are caused by multiple etiologies and abnormal immune response,with the characteristics of recurrent chronic nonspecific intestinal inflammation.Large amount of research believes that IBD is a pathophysiologic consequence of excessive and uncontroled inmune response to normal intestinal mucosa and flora.Some study indicates that allicin could mudulate immune response.Allicin was used to intervene in TBNS induced colitis model in this experiment to study its treatment effect and the possible mechanism related to IL-6/JAK/STAT3 pathways.Methods:50 SD rats were divided into five groups randomly:①blank control group,②TNBS model group,③allicin treatment group,④mesalazine treatment group and ⑤allicin plus mesalazine treatment group.ALL groups except for blank control group were intervened by 5%TNBS plus 50%ethanol solution to make acute colitis models,allicin and mesalazine were used to intervene in treatment groups(allicin 30ng/kg;mesalazine 30mg/kg);rats in TNBS group and blank control group were gavaged with noraml saline of the same dose for ten consecutive days,during this period rats mental status,hair,food intake and activity condition were observed each day.Rats body weight changes were recorded every day,stool charcters and bloody stool were oberved andstandard DAI scoring were carried out.Rats were killed after intraperitoneal anesthesia,blood sample and colon were observed and tested;all rats colons were observed for pathological changes by naked eyes and were observed under microscope after HE staining and colon histological scores were carried out.Levels of IL-6 and IL-10 in rats serum were assayed by means of ELISA,expressions of STAT3,SOCS3 in rats colon tissues were assayed by immunohistochemistry and were counted.STAT3 and SOCS3 mRNA expressive quantity were assayed by RT-PCR.Experiment data were analyzed by statistical software and result was gained.Results:1.Compared with blank control group,index like rats DAI scores,colon histological scores and weight in each treatment group and TNBS model group are of significant statistical differences,which indicates the success of model copy;2.Rats serum IL-6 levels assayed by ELISA are of significant statistical differences between TNBS model and the other three treatment groups(p<0.05),on the average of IL-6 levels,it is the lowest in allicin plus mesalazine treatment group,which indicates that allicin can inhibit secretion of inflammatory factors,while allicin plus mesalazine have the highest inhibition of inflammatory factors secretion.3.Ten days after intervention,immunohistochemistry shows that in terms of the numbers of positive cells in colon tissues,numbers of positive cells with expression of STAT3 are the largest in TNBS model group,and the smallest are in the blank control group,among treatment groups,expressions of STAT3 are of the smallest numbers in allicin plus mesalazine treatment group,and the difference is of statistical significance compared to allicin treatment group.n terms of counting of SOCS3 positive cells,in TNBS model group,the numbers are of statistical significance compared to the rest four groups(p<0.05);among treatment groups,there are the largest numbers of SOCS3 positive cells in allicin plus mesalazine treatment group,and the difference is of statistical significance,compared to allicin treatment group.STAT3 and SOCS3 in colon tissues in blank control group have weak expressions;STAT3 expressions in TNBS model group are obviously higher than those in treatment groups,the difference is of statistical significance(p<0.05);SOCS3 expressions in TNBS model group are obviously lower than those in treatment groups,the difference is of statistical significance(p<0.05).Conclusion:1.TNBS/ethanol induced rat colitis model is successfully constructed in this experiment;2.Mortality rate,weight change,histopathological change and serological assays are different among each group,allicin administrated by gavage has a certain therapeutical effect on IBD model,allicin combined with mesalazine have better treatment effect than that of allicin or mesalazine which is used alone;3.Allicin may inhibit the secretion of IL-6 and inhibit the expressions of IL-6/JAK/STAT3 pathways to reduce colon inflammation. |
