Liposarcomas are malignant soft-tissue tumors that originate from mesenchymaltissue. Many of these tumors often occur in the deep soft tissues and retroperitoneum ofan adult person. Liposarcoams in the retroperitoneum have a tendency to relapse aftersurgeries because of thedeep tumor location, which often makes it difficult to get aclean surgical margin. For retroperitoneal liposarcoma, there are no effective drugsexcept surgeries. Curcumin is a natural phenolic substance extracted from the CurcumaLonga Linn. Curcumin has a wide spectrum of pharmacological effects, like antioxidant,anti-inflammatory, anti-atherosclerotic and anti-tumor. Previous studies found curcumincould inhibit the growth of SW872, but its mechanism was not completely clarified.Inour present study, we hope to further explain the anti-tumor effects and its relatedmechanism of curcumin on the liposarcoma cell line SW872.In vitro, the anti-tumor effects of curcumin with different concentrations on SW872cells were detected by MTT method. The flow cytometry was used to determine theeffects of cell cycle distribution and apoptosis of SW872cells treated by differentcurcumin concentrations. The gene expressions of Bad, Bcl-2,TNF-?and Survine ofSW872cells treated by different curcumin concentrations were detected by usingreal-time PCR. Finally, the anti-tumor effect of curcumin on liposarcoma in vivo wasmeasured by using SCID mice's subcutaneous xenotransplanted tumor models.In vitro, the MTT results showed the inhibition rates of curcumin to SW872cellsgradually increased with the increase of drug concentration of curcumin, and the50%inhibitory concentration (IC50) was7.1±0.4?g/ml. The flow cytometry results showed,compared with control group, the apoptosis rates were higher in in experimental groups,and more SW872cells were blocked in G2/M phase. With the increase of drugconcentration of curcumin, the expression of gene Bad increased(P<0.05), while that ofgene Bcl-2decreased(P<0.05), and expressions of gene TNF-? and Survine were not affected. In vivo, curcumin could inhibit the growth of subcutaneous tumors of SW872cells. Compared with control group, the tumor inhibition rate of experimental group was62.5%(P<0.05).Curcumin had high efficiency and low toxicity, and the body weightdifferences between control and experimental groups in the beginning and at the end ofthe experiment got no statistical significance(P>0.05).To sum up, curcumin can inhibit the growth of SW872cells both in vitro and in vivoand may be used as a potential agent to treat liposarcoma through inducing apoptosis,which is worth further research.
