Objective:Exploring mechanism of Wnt/?-catenin signaling pathway in the paclitaxel treatment of cervical cancer..Methods:1. To detect with different density of paclitaxel on the proliferation of human cervical carcinoma Caski cell by MTT colorimetric assay.Collecting of cervical cancer Caski cells in logarithmic growth phase, addining 100?l different concentrations of paclitaxel after the cells adherence, the drug density in each group were 0.001?g/ml, 0.01?g/ml,0.10?g/ml, 1.00?g/ml, 10.00?g/ml, 100.00?g/ml,1000.00?g/ml, the each density set 5 Double-Pored. After incubated for 48 hours, each Pore was added 20?l 0.5% MTT solution, continuing cultured for about 4 hours, masuring the absorbance of each Pore (OD) in OD570nm by ELIS A calculated inhibition ratio and IC50.2. Affect paclitaxel protein expression of ?-catenin in Caski cell.Coverslips placed in 24-well plates hole again Caski cells were cultured in the experimental group added paclitaxel (final concentration 6.04nmol/L), paclitaxel control group were not treated incubator 48h incubation,?-catenin protein expression were detected by immunohistochemistry. Caski cell were observed, the positive rate is calculated.3. Effect of paclitaxel on c-myc expression of Caski cells in mRNAAfter cultured Caski cells, paclitaxel concentration 6.04nmol/L put in experimental group, and equal volume of culture medium put in control group. After 48 hours. RT-PCR was used to detect the c-myc gene expression in each cell.Results:1. Affect of paclitaxel on expression of ?-catenin protein in Caski cell.According to experimental results, the experimental group and the control group of ?-catenin protein were expressed in the cytoplasm and nucleus, the control group is more dense brown granules masslike. In experimental group, the positive rate was 53.3%. In control group, the positive rate was 71.7%. The positive rate in experimental group was significantly decreased (P<0.05).2. Effect of paclitaxel on the expression of c-myc in mRNA.The experimental group and the control group can be detected the c-myc mRNA expression, mRNA contents in experimental group decreased significantly compared with the control group (P<0.05)Conclution:Paclitaxel can inhibit the expression of-catenin protein in cervical cancer Caski cells, thereby blocking the wnt/?-catenin signal transduction pathway, Down the expression of c-myc gene, and inducing apoptosis of cervical cancer Caski cells.
