Objectives:To investigate the association between Pro200Leu gene polymorphism of Glutathione peroxidase-1(GPx-1) and type2diabetes mellitus and diabetic peripheral neuropathy in Han Chinese in Changde region in Huan Province.Methods:The study selected patients diagnosed with type2diabetic peripheral neropathy(DPN (+) group,n=150)?type2diabetes mellitus (DPN (-) group,n=150) and normal control subjects(NC group,n=265). Exmined their clinical parameters.And extracted DNA? PCR amplified target gene.The genetypes of Pro200Leu polymorphism in GPx-1gene in those individuals were determined by PCR-direct sequencing. The genotype and allele frequency distribution of Pro200Leu gene polymorphism were compared in the NC group and T2DM group.The genotype and allele frequency were also compared in DPN(+)group and DPN(-)group.Results:?There were significant differences of genotypic and allelic frequencies of Pro200Leu polymorphism in GPx-1gene between T2DM group and NC group(P<0.001; P<0.001).The OR for T2DM was2.154for genotype CTvsCC after adjustment for age?sex and BMI.? No significant difference of genotype and allele frequence distribution between DPN(+)group and DPN (-) group (P=0.079; P=0.065)Conclusions:?Pro200Leu polymorphism of GPx-1gene was associated with the onset of type2diabetes in Han population of Changde region in Hunan Province,T allele may be a risk factor of T2DM.?There was no association between Pro200Leu polymorphism of GPx-1gene and DPN.
