Objective:We studied the mechanism and effects how fig leaf extract is protective in homozygous Osteoprotegerin (OPG) knockout mice (OPG-P-mice) to against osteoporosis, with expression characteristics of interleukin-1(IL-1) and tumor necrosis factor-a (TNF-a).Methods:60female mice were gained with eight-week-old non-frequent breeding law, then randomly divided into three groups. Group A:the experimental group, Group B:control group, and Group C:blank group, each group has20mice.10OPG mice in each group were killed after one month and two months medication. HE staining method was employed to measure the number of trabecula, osteoblasts and osteoclasts in each group, meanwhile IL-1and TNF-a expression levels from left femur OPG mice were detected with immunohistochemistry methods.Results:There is no significant of bone trabecular morphology, osteoblast number and osteoclast number among experimental group, control group and blank group after been gavaged30days later with HE staining (P>0.05), and difference can be found between experimental group, control group and blank group on60th day (P<0.05), while the difference was not found between experimental group and control group (P>0.05). There was no significant difference of IL-1and TNF-a positive expression cells number with immunohistochemical staining after30days of gavage among experimental group, control group and blank group (P>0.05). While the difference can be found between the experimental group to control group, as well as to blank group after gavaging for60days, which was characterized with decreasing of IL-1and TNF-a positive cells number (P<0.05). But the difference can not be found between the experimental group and control group (P>0.05).Conclusion: Our results offer evidence that fig leaf extract mediates anti-osteoporosis in the OPG-P-mice primarily via activation of the osteoprotegerin.
