| Plant pathogenesis-related proteins(PRs)is a kind of protein induced by biological or abiotic stresses.Among them,PR-5 is a kind of active protein with antifungal and abiotic stress activity.Protein PR-NP24 is a special protein in the PR-5 family.The specific expression of protein PR-NP24 in this study,expression patterns in plant fruit,induced expression in adversity and related genes in the fruit of Solanaceae downy ground cherry.The results of the study are as follows:(1)The tissue specific expression and mass spectrometric identification of PR-NP24 in different varieties of tomato fruit showed that PR-NP24 had different tissue specific expression differences in the fruit of Micro-Tom and the fruit of common varieties.Among them,the outer epidermis has the highest expression level,while the outer epidermis has relatively low expression level.(2)The expression of PR-NP24 in Tomato under abiotic stress was studied.It was found that the expression of PR5-NP2 in the outer epidermis of tomato did not change significantly under cold storage.Sodium Chloride Solution can significantly induce the up regulation or down regulation of PR-NP24.However,different forms of external injury did not significantly affect the expression of PR-NP24 in tomato.(3)It was found that the expression of PR-NP24 in the inoculation site was significantly down,and the expression of the tissue was downregulated at the inoculation site.Pretreatment of tomato with salt solution and inoculation test showed that induction and regulation of PR-NP24 expression affected tomato resistance.(4)Through the comparison and screening of PR-NP24 protein in the database,the fruit of Solanaceae and Solanaceae was analyzed and the gel expression was detected.It was found that some proteins with high homology with protein PR-NP24 were found in some plants.Among them,the expression level of Physalis pubescens is the highest in Solanaceae.(5)According to the above results,the PR-NP24 homologous gene PpTLP1 in the hair acid pulp was cloned and bioinformatic.It was found that the protein had a transmembrane helix,a signal peptide,and belong to GH64-TLP-SF superfamily,and was located extracellular.In particular,it has a conserved domain of thaumtin or thaumatin-like protein,and there are four secondary structure at the same time.In addition,PpTLP1 protein in berry fruit is also characterized by tissue specific expression.The pathogenesis-related protein PR-5 protein has antibacterial and anti abiotic stress activity.Through the above studies,it has been found that protein PR-NP24 has the specificity of tissue expression and its expression is affected by adversity.The protein is widely found in plant fruit(such as dowmy ground cherry).This study provides a theoretical reference for the in-depth study of the expression of PR5 and its regulation mode,and the induction of PR-NP24 expression in order to improve the resistance of plants,and also provides new ideas for the transport and storage of tomato. |
