Sanguinarine is a benzophenanthridine alkaloid,mainly for anti-inflammatory and promoting the growth in the production.To study the therapeutic effect and mechanism of sanguinarine on layers salpingitis,it will provide the test basis for the treatment of sanguinarine on layers salpingitis,we conducted the following tests:1.Screening of modeling methods on layers salpingitis: 10 layers were randomly divided into 3 groups.E.coli and phenol mucilage were injected into the oviduct,and control group was set.The clinical manifestations and pathological changes were observed,and the histological changes in tubal slices were observed.Results: The inflammatory lesions of the phenol mucilage group were obvious,and the effect of inflammation was significantly better than that of Escherichia coli group.2.Investigating of modeling methods on layers salpingitis: 10 layers were randomly divided into modeling group and control group,each group of 5,respectively,with phenol mucilage and saline into the oviduct,after 48 h 2mL blood per layer was collected to measure the contents of IL-2,IL-6,TNF-? and IFN-? in serum,3 layers of each group were randomly collected oviduct for visual observation and microscopic examination.Results: The inflammatory reaction was observed in the naked eye and microscopic examination of modeling group,and the contents of IL-2,IL-6,TNF-? and IFN-? in the serum were significantly higher than those in the control group.3.Prevention test: 36 layers were randomly divided into prevention ?,?,? groups(test drug: sanguinarine),amoxicillin control group,modeling group and control group,each group of 6.The experimental groups were treated continuously for 14 days,the modeling group was only modeling with no drug,the above groups were modeled at 8th day,and the control group was not treated,and at 15 th day 3 layers of each group were randomly collected oviduct for visual observation and microscopic examination.Results: There were no obvious clinical symptoms in the prevention ?,?,? groups and the amoxicillin group.The inflammatory reaction by visual observation and microscopic examination was slighter than that of the modeling group.4.Treatment test and study on the mechanism: 36 layers were randomly divided into treatment A,B,C groups(test drug: sanguinarine),amoxicillin control group,modeling group and control group.Start treatment after modeling 48 h,at 8th day 3 layers each group were collected 2 mL blood to mesure the contents of IL-2,IL-6,TNF-? and IFN-? in the serum,and two copies of oviduct were collected,one for visual observation and microscopic examination,one for the determination of inflammatory factors expression.Results: There were not inflammatory lesions by visual observation and microscopic examination in the treatment A,B,C groups,the contents of IL-2,IL-6,TNF-? and IFN-? in the serum were lower than those in the modeling group,and the levels of IL-2,IL-6,TNF-? and IFN-? were also lower than those in the modeling group,the effect of treatment A group(17.75 mg)was the best.Conclusion: The phenol mucilage method is an ideal method for modeling salpingitis in layers;3.75 mg sanguinarine has a good preventive effect on layers' salpingitis,while 17.75 mg sanguinarine has a good therapeutic effect,the treatment mechanism may achieve through the down-regulation of IL-2,IL-6,TNF-? and IFN-? gene expression.
