Yak is one of the major livestock species distributed in the alpine regions of the Qinghai-Tibet Plateau and its surrounding areas,which provides abundant milk,meat,wool and other livelihoods for the herdsmen.For the high altitude living conditions,the physiology of yak including growth,metabolism,and reproduction has well adapt to the local environment,and which has being an ideal animal model for the study of the hypoxia adaptation mechanism of plateau animals.HIF(Hypoxia Inducible Factor)is an important transcription factor of the oxygen signal transduction and plays an vital role in response to hypoxia.In order to investigate the expression characteristics of the HIF-1? gene and the encoded protein in the reproductive organs and placenta development of adult female yak,which further understand the adaptive mechanism of yak in hypoxia environment,the ovaries,oviducts,uterus and placenta tissues of early pregnancy were collected from 5~8 years old yaks to detect the expression levels of HIF-1? gene and the encoded protein by using real-time quantitative PCR and immunohistochemistry.1.Results showed that the expression of HIF-1? mRNA was decreased in ovaries,oviducts and uterus tissues during estrus,While and during pregnancy,expression HIF-1? mRNA in placenta tissues was significant higher than in oviducts(P<0.05).Moreover,significant differences of the HIF-1? mRNA level were found in the ovarian tissues at different stages,which showed the highest expression at luteal phase,but with a lower expression during pregnancy.In addition,the HIF-1? mRNA level in oviducts during pregnancy was apparently higher than that in the luteal phase(P<0.05),and also with a peak in the uterus during pregnancy.2.Results of immunohistochemistry revealed that the HIF-1? protein positive reaction areas were mainly distributed in epithelium of ovarian,granular luteal cells,granule layer of follicle,mucosal epithelium of fallopian tube,endometrial epithelium,uterine gland and epithelium of fetal chorion.Collectively,the expression HIF-1? shows a time-space differential expression pattern in the major reproductive organs of adult female yaks.The higher expression of HIF-1? in ovarian tissues during estrus and placental tissues during pregnancy suggests that the roles of these organs played at their corresponding period and functions in the enhancement of metabolism are related to the hypoxia adaptation mechanism mediated by HIF-1?.And these results provide basic information for further study of the mechanism of hypoxia adaptation mediated by HIF-1? in mammalian reproductive regulation and reproduction of plateau animals.
