Tiger pufferfish?Takifugu rubripes?is a kind of high-grade precious fish specie,with high nutritional value and economic value.It gradually become one of the most promising species in aquaculture industry of China,because of the opening of the striction of tiger pufferfish since.2016.In the present study,tiger pufferfish?male 424.7±45.3 g,female 436.8±62.8 g?from same batch were randomly divided into two groups,one group in recirculating aquaculture system?RAS?,while another group in off shoresea cage aquaculture system?OSCS?,for 70 days breeding experiment.and studied the two mode The difference between male and female from two modes was studied in the present research,the experiment contents were as follows.First,the hepatopancreas somatic indices,gonad somatic indice,viscera somatic indice,and condition factorand specific growth rate were analysed by measuring body length,body weight,liver weight,liver weight and gonad weight of tiger puffer.And the mRNA expression levels of growth factors in the tiger puffer liver,like as igf1,igf2,igfr1,igfr2,ghr1and ghr2 gene,were determinated by RT-PCR method.The results showed that the transcription level of growth-related genes of female fish in OSCS was significantly higher than fish culturedin RAS,while no significant difference was found in the ratio of hepatosomatic index,viscerosomatic index and gonadosomatic index.These reults indicated that the growth performance of the female tiger puffer culture in the OSCS is more superior than fish cultured in RAS,and the female tiger puffer culture in the OSCS obtained the best growth performance.Secondly,the difference of blood physiological and biochemical indexes of male and female tiger puffer between two aquaculture modes was identified by analyzing the level of serum glucose,cholesterol,triglycerides,high-density lipoprotein cholesterol,low-density lipoprotein cholesterol,albumin,globulin,sodium,potassium.The results showed that there was no significant difference in the level of blood glucose,serum total cholesterol,triglyceride,low density lipoprotein,and white pellets between male and female tiger puffer in different culture systems.The level of serum high-density lipoprotein,total protein,albumin,globulin,Na+,Cl-,Ca2+of males tiger puffer cultured in the OSCS were significantly higher than female tiger puffer cultured in the OSCS and fish cultured in RAS?P<0.05?.Thirdly,the antioxidant and metabolic capacity of the tiger puffer liver was analysed by determining the activity of hepatic anti-oxidant enzymes?SOD,MDA,CAT and GSH-PX?,metabolic-related enzymes?AST,ALT,LZM,PK,Na+-K-/ATP?,antioxidant factor?cat,cu/zn-sod,mn-sod?and the mRNA expression levels of stress proteins?hsp70 and hsp90?.The results showed that the activities of hepatic SOD,CAT and GSH-PX in male tiger puffer which was cultured in OSCS were significantly higher than fish cultured in the RAS?P<0.05?,whereas AST and ALT activities remained constant?P>0.05?.Meanwhile,similar results were found in the mRNA expression of hepatic sod and hsp70 and hsp90 where OSCS group was significantly higher than RAS group.These results indicated that the hepatic anti-oxidant and physiological metabolism ability of fish in OSCS group was higher than RAS group,where the male tiger puffer cultured in OSCS achieved the best antioxidant and metabolic capacity.Finally,the mRNA expression of apoptosis-related genes?bax,bcl2,p53,caspase3caspase8 caspase9?and immune-related factors?tnf,baf,il-6,il-7,il-12,c3?in the liver,spleen and head kidney were evaluated.Results showed that the hepatic expression of caspase3 and caspase8 mRNA of male tiger puffer cultured in the OSCS was significantly higher than that in RAS,whereas the mRNA level of p53 and caspase9 in male tiger puffer cultured in the OSCS was significantly lower than that in RAS and female tiger puffer cultured inOSCS.The mRNA expression level of the immunity-related factor il-6 and c3 in male tiger puffer cultured in the OSCS was significantly higher thanfish cultured in RAS?P<0.05?.These results suggested that the anti-apoptosis and immunity ability of the male tiger puffer cultured in OSCS was better than fish cultured in RAS.In conclusion,these results indicated that the growth performance,metabolic state of the female tiger puffer reared in OSCS were better than those in RAS,whereas the capacity of antioxidant,anti-apoptosis and innate-immune in the male tiger puffer reared in OSCS were better than those in RAS.Thus,the OSCS was more suitable than the RAS for the short-term culture of subadult tiger puffer.These findings will provide the basic data for management of tiger puffer and enhance our understanding of the combined land?RAS?and sea?OSCS?alternative aquaculture model of tiger puffer in captive.
