Lablab purpureus Sweet is a genus of leguminous lentils,which is both edible and medicinal.It has been widely distributed since ancient times.Dolicho LablabL are commonly used Chinese herbal medicines,and the "Compendium of Materia Medica" records: Make up the five internal organs,Cure vomiting;Long-term use of hair is not white,cure a cold,treated with women;Relieve alcoholism,relieve dolphin fish poisoning;stop phlegm,relieve summer heat,warm the spleen and stomach,Remove hot and humid,stop thirst.At present,the research on the molecular biology of Lablab purpureus is still in its infancy,and no studies have been reported on the construction and localization of medicinal Dolicho LablabL.In this experiment,medicinal Dolicho LablabL(abbreviated as W)and their mutant YuCuibian-1(abbreviated as R)were used as parent materials,and 143 F2 of W × R were used as mapping groups.Genetic linkage maps were constructed using SLAF-seq.According to the transcriptome data of YuCuibian-1,a new Indel marker was designed and developed,the SLAF map was marked and encrypted,the annotation on the above map was functionally annotated.Investigate F2 stalk color,leaf color,flower color,flower color,alfalfa pigment,alfalfa size,alfalfa crispness,angle bend,grain color,alfalfa length,alfalfa width,thin edge thickness,thick edge thickness,total weight,Net weight,grain weight and dry weight 17 traits.And QTL mapping for 17 traits was performed using WinQTLCart 2.5 software.The main results are as follows:1.Using the SLAF-seq technology,we conducted high-throughput sequencing of 143 individuals in the F2 population of medicinal Dolicho LablabL,identified SNPs,and constructed high-density genetic maps of Lablab purpureus with SNP data.The map contains 2193 SLAF markers and 11 linkage groups,the total length was 1414.18 cM with an average distance of 0.75 cM.2.The above figure was compared with Unigene in the Lablab purpureus transcriptome and 914 functional marker genes were successfully annotated.3.According to the transcriptome sequencing data,the SLAF map was encrypted,80 pairs of Indel markers were screened,and 28 Indel markers were added.The average map distance was reduced from the original 0.75 cM to 0.6cM.4.QTL mapping was performed on 17 traits of medicinal Dolicho LablabL.A total of 29 QTLs were detected and distributed in 25 marker regions of 7 linkage groups.The number of QTLs detected by individual traits was 1-3,and the individual QTL accounted for 1.46%-36.39% of the phenotypic contribution,of which 25 were major QTLs,covering all traits with phenotypic variation of 10.05%-36.39%.A single major effect QTL was detected in eight traits,including flower color,flower color,amaranth pigment,axillary width,thickness of thin edge,thickness of a thick edge,axillary weight and axillary dry weight.There were 2 QTLs in six traits,including leaf color,pod size,glutinous crispness,grain color,pod length,and grain weight.The 3 QTLs detected by three traits,including stalk color,angle bending,and total weight.5.There were 161 molecular markers in the 29 QTL markers covered by the 17 traits,46 were markers for functional annotation using the information of the transcriptome of YuCuibian-1.It is worth mentioning that Marker 110224 was located on the 7th linkage group,is in the main QTL interval with overlapping stalk color and soybean meal size,is a molecular chaperone for post-translational modification;the Marker 12680 was located on the 4th linkage group,is in the main effect QTL interval with overlapping the flower color and the seed color overlap,it is similar to the plant protein transposase in the tobacco and kidney bean genomes.
