In order to investigate the genetic laws of fruit spine base size in cucumber,a few suitable populations for locating related genes in cucumber was established and selected. In this study, the development process of fruit spine base size was firstly observed and counted, which the results showed that the day of flowering was the best time for phenotype identification in fruit spine base size.Secondly, three parental materials with different fruit spine base size were used to construct two genetic groups and major gene plus polygene model was used for genetic analysis.Genetic model analysis showed that the fruit spine base size in first population with CNS5 and RNS4 were consistent with the C-0 model, the additive-dominant-epistatic polygene model.Multi-gene additive and dominance effects were positive, and epistatic effects accumulated positively. The heritability polygene in F2 population from two seasons in autumn 2016 and spring 2017 were 79.21%and 71.25%,environmental effects were 20.79%and 28.75%,respectively.In the second group from the RNS4 and the CNS16,The results showed that fruit spine bases size was consistent with the E-2 model, two pairs of additive-dominant major genes plus additive-dominant polygene. In the F2 population, the heritability of the major gene was10.42%and the polygene heritability was 59.54%,the total heritability was 69.96%.However,the phenotypic value of BC1F1 from the second group was continuous distribution, and from which the plants of extreme and medium were selected to continue further backcrossing with CNS16,as a recurrent parent, and expected to selection appropriate presence of segregation ratio populations from the BC2F1 generation or continued backcrossing for mapping the fruit spine base size related genes. Moreover, some molecular markers were screened between differential materials from our laboratory will be used to mapping analysis. The results of this study laid a foundation for breeding and mapping, and which would be helpful for further exploration of fruit spine base size related genes in cucumber.
