| Mangrove ecosystems rank fourth in the world for high service value ecosystems.Mangroves have many special capabilities,such as adsorption of harmful substances,it attracted the attention of the vast number of scientific researchers.It is expected that the relationship between mangrove microbial diversity and environmental homeostasis,as well as the function of microorganisms,can be obtained through the study of environmental microbes in mangrove forests.In this study,eutrophication of Xiamen under the condition of two rivers between mangrove soils were sampled from four sites of distance,the TN,TC,TS,TP,EC,Water,content,pH,physical and chemical properties were detected,with the total amount of soil microorganism and soil microbial analysis of interaction between pure culture.By the way and in soil bacteria,actinomycetes were isolated,and explore its diversity.Through metagenomic taxonomic determination,detection of species and abundance of soil microorganisms and the prokaryotic eukaryotic organisms.In contrast to pure cultivation and free of cultivation of two methods,comprehensive analysis of soil microbial diversity.Using metagenomic genome the detection method,analysis of functional genes of mixed soil samples.300 strains of bacteria and 75 strains of actinomycetes were isolated from four soil samples by pure culture.Among them,3 phyla and 6 genera of bacteria,2 phyla and 5 genera of actinomycetes were isolated at P1 site.3 phyla and 7 genera of bacteria,1 phyla and 3 genera of actinomycetes were isolated at P2 site.1 phyla and 3 genera of bacteria,2 phyla and 3 genera of actinomycetes were isolated at P3 site by 16 s rRNA.A total of 5 phylum and 15 genera of bacteria were isolated from the four samples,and 2 phylum and 7 genera of actinomycetes were isolated from the four samples.The dominant bacteria group was Bacillus sp.(82.12%).The dominant flora of the soil actinomycetes was Micromonospora(72%).The bacteriostatic activity of 18 actinomycetes strains was screened out by using the fermentation broth of strain,in which 12 strains of Micromonospora(66.67%),4 strains of Streptomyces(22.22%)and 2 strains of Actinomadura(11.11%)were found to have antimicrobial activity.Using macrogenomic taxonomy,Phylogenetic analysis of prokaryotes showed that there were 40 phyla,67 classes,89 orders,179 families,499 genera in P1,38 phylums,67 classes,88 orders,168 families,491 genera in P2,41 phyla,71 classes,92 orders and 171 families 814 genera in P3.There are 39 phylum,71 classes,96 orders and 183 families 482 genera in P4.Proteobacteria is dominant genus of four soil samples(50.26%-52.53%).Desulfomonile is the the dominant genera(3.46%-4.07%).The diversity and richness of prokaryotes in soil samples at four loci are basically similar.There are 31 phyla,45 classes,61 orders,61 families,186 genera and 186 genera in the samples of eukaryote P1.There are 32 phyla,46 classes,56 orders,69 families,177 genera,including 32 phylums,43 classes,52 orders and 62 families.There are 34 phylum,47 classes,54 orders,5 families and 163 genera in 186 genera of P4.31 samples,45 classes,61 orders,61 families,and 186 genera in P1 samples;32 samples in 46 samples,56 orders,and 69 families in P2 samples.177 genera;32 samples,43 classes,52 orders,62 families,186 genera in P3 samples;34 samples in P4 samples,47 classes,54 orders,5 families,163 A genus.The eukaryotic microorganisms were dominated by Apicomplexa and Malassezia(11.18%-39.84%)was the dominant genus.By macrogenomics genome-wide analysis,the gene set protein sequence was compared with COG database to classify functional genes into energy production and conversion.There were 49 antibiotic genes in antibiotic resistance gene alignment.The highest value was macrolides,followed by Bacteriopeptide and Bacillus subtilis antibiotics.Through carbohydrate activity enzyme comparison,the quantity of active enzymes was analyzed.The order from high to low is glycoside hydrolase > carbohydrate ester lyase > redox enzyme > carbohydrate binding domain > polysaccharide lyase.Component,the biological process involved in three aspects.Through the SEED database comparison,Functional genes were classified into 28 specific classifications,such as carbohydrates. |
