Foot-and-mouth disease was an acute,febrile,highly contagious zoonosis caused by the foot-and-mouth disease virus which had great harm to livestock production and been listed as the head of"A"class infectious disease.Outikang was a compound Chinese herbal medicine developed under the guidance of the theories of Chinese Veterinary Medicine to enhance the body's immune function and strengthen the immune effect of the foot-and-mouth disease vaccine.The experiment respectively studied the impact of the medicine on immunological function of mice and on antibody titer of foot and mouth disease vaccine type O,type A and type Asia?,serum levels of ten cytokines and biochemical indices in cows and the acute and subchronic toxicity of Outikang.The results were as follows.1.Impact on immunological function of mice:Outikang could significantly rise the immune function of erythrocytes,and there was a positive correlation between effect and dose.The medicine could increase the neutrophil swallowing rate and the abdominal cavitymacrophage swallowing rate,but had no obvious effect on phagocytic index.The number of plaque-forming cell increased significantly and there was a positive correlation between effect and dose.The percentage of E rosette-forming cells improved significantly.The medicine could promote the proliferation of mouse spleen lymphocytes significantly.2.Impact of Outikang on antibody titer of foot and mouth disease vaccine?FMDV?type O,type A and type Asia?,serum levels of ten cytokines and biochemical indices:there were no significant differences in antibody titer of FMDV between normal control group and drug group on 0 d and 56 d after vaccination?P>0.05?,while on 14 d the effect of Outikang on antibody titer of FMDV reached very significant level?P<0.01?and on 28 d the difference was significant?P<0.05?.Compard with normal control group,the contents of T-Bil in drug group was significantly decreased on 0 and 28 d?P<0.05?,while it was extremely significantly decreased on 56 d?P<0.01?.The contents of Glu had significant difference on 28 and 56 d?P<0.05?.The contents of P had extremely significant difference on 0 d?P<0.01?,which was within the normal range.On 0 and 14 d after vaccination,the content of CD4 significantly increased in the drug group comparing to those in normal controls?P<0.05?.On 28 d after vaccination,the content of CD4,IL-2,IL-6,IL-10,IL-12 and IFN-?extremely significantly increased in the drug group comparing to those in normal controls?P<0.01?.On day 56 after vaccination,the contents of IL-6 and IL-12 extremely significantly increased in the drug group comparing to those in normal controls?P<0.01?,while the content of CD4,IL-2,IL-4 and IL-18significantly increased?P<0.05?.3.Result of acute toxicity and maximum dosage study:Outikang had no acute toxic effects in mice.The LD50 of the medicine was more than 76.83 g·kg-1.The maximum dosage was more than 120 g·kg-1.4.Results of subchronic toxicity:80 healthy adult SD rats were averagely divided into 3 drug groups?high-does group,middle-does group and low-does group?and a normal control group.The drug groups were given the medicine for 28 days running.The result of blood routine and biochemical test showed that the medicine has no obvious harm to liver function,kidney function and cardiac function.The observing pathological section showed that the histological structure of organs had no obvious injury.Therefore,Outikang had no obviously toxic side effect and improved the immune function of mice.It could significantly rise the antibody titer of FMDV type O,type A and type Asia?.While it had no damage to the liver and kidney functions and had cholagogue effect and enhance cellular immunity and humoral immunity function obviously.The study also provided a scientific basis for the clinically widely using of the medicine.
