Identification Of Begomoviruses In Hunan Province And Hainan Province

Virus in the family Geminiviridae are plant viruses with a single stranded DNA,which were spread by Bemisia tabaci or leafhoppers,and the host range of which were very extensive,causing devastating damage to crops in the areas of tropical and subtropical.In recent years,geminiviruses continue to expand,and they have been found in Yunnan,Chongqing,Hainan,Guangxi,Shandong and many other provinces,therefore,early detection of geminiviruses has become more and more important.In order to further determine the distribution of geminivirus in China,samples collected from Hunan and Hainan were detected and identified.In 2016,six plant samples were collected from Hunan Province with symptoms of suspicious geminivirus infection.After extracting the total DNA,we used them as templates for rolling circle amplification(RCA).Then enzyme digestion on RCA products was carried out.Sequencing results revealed that the isolates from tomato and morning glory shared 99% of homology with tomato yellow leaf curl virus(TYLCV)and sweet potato leaf curl virus(SPLCV),respectively.At the same time,we used universal primers beta01/beta02 and PCRc1/PBL1v2040 to amplify,but DNA-B or DNA-? were not been detected,these results indicate that tomato and morning glory isolates we detected were monopartite geminiviruses.Both of them encoded 6 ORFs: AV1 and AV2 in the virus strand,AC1,AC2,AC3 and AC4 in the complementary strand.This is the first report of geminivirus infection in Hunan Province.In 2016,the tomato plants with symptoms of suspicious geminivirus infection were collected from Hainan Province.Based on the RCA-DNA technology,we used universal primers BegoAFor01/BegoARev1 and beta01/beta02 to amplify,results showed that the tomato isolate shared 99% of homology with tomato yellow leaf curl China virus(TYLCCNV).The satellite DNA-? sequence was also been detected.This is the first report of TYLCCNV infection in Hainan Province.
