At present,most Photosynthetic Bacteria is liquid form.Existing problems are easy loss,inconvenient preservation and transportation.The solution to above problems is attaching the Photosynthetic Bacteria with suitable carrier,making preparation solid state.The study intends to explore improvement of photosynthetic bacteria biomass and preparation of solid agent.to Investigating the purification effect on artificial aquaculture water through the experiment.The main contents and results of research are as follows:1.Purifying and identifying the photosynthetic bacteria.Purifying strains from Photosynthetic bacteria flocculating agent,which is from Laboratory original open culture.By observing the colony and cell morphology,sequencing 16 s rDNA,and Constructing of the evolutionary tree,we can finally determining that the Isolation and purification of photosynthetic bacteria strains are Class ball red bacteria.2.Mixed culture and application research on Photosynthetic bacteria and bacillus.In this paper,orthogonal test natty and photosynthetic bacteria mixed culture inoculat-ion and liquid volume of agents when mixed culture,make photosynthetic bacteria reached the maximum number of viable cells in the shortest possible time.The results showed that:20%of photosynthetic bacteria inoculation,natty inoculums 2%,agent’s liquid volume 60%/250 mL flask culture best.At this point the number of photosyn-thetic bacteria reachs 22.8 billion/mL.It is three times of a pure bacterial culture of photosynthetic bacteria.Mixed Photosynthetic bacteria and bacillus natto,exploring the impact of different amounts(0.1%,0.5%,1%,3%,5%)of mixed bacteria on aquaculture water purification.The results show that the optimum mixing ratio of two kinds of bacteria is 1:1(V:V).At this scale,treated aquaculture water 48h,various additions on nitrite degradation rate reached 90%,while the degradation rate of ammonia are 56.4%~-15.7%.Showed the influnce of different inoculation of agents for N-nitroso degradation is small,greater impact on the degradation rate of ammonia.3.Preparation and preservation to PBS-curing agents.The experiment adopts direct adsorption method and the adsorption of the oscillation.Respectively,three kinds of support that is charcoal,ash,activated carbon(bibulous rate is 39.33%、18.67%、222%)are prepared by solid-type.① Direct adsorption method:according to the carrier bibulous rate,we can add the photosynthetic bacteria mixed culture with photosynthetic bacteria and natto preparation into adsorption agents.The result shows that the direct adsorption agents of charcoal,ash,activated carbon to the release rate of photosynthetic bacteria are 21.77%、5.58%、76.25%;the water content of each adsorbent agents are 15.65%、4.8%和 27.5%;the highest number of viable cells of photosynthetic bacteria in each bacterium are 2.7 billion/g,0.57 billion/g and 78 billion/g.The experiments on preservation of each adsorption agents show that the storage time for photosynthetic bacteria in charcoal and activated carbon is 60 d.②Oscillation adsorption:the experiment shows it has better effect in this condition:Charcoal and activated carbon 10g/100ml,Coal ash 30g/100ml,Oscillation speed 120r/min,Oscillation time 30min.The moisture get by Oscillation adsorption:Charcoal 3.2%,Coal ash 1.955%,activated carbon 11.96%.The most significant number of viable cells of photosynthetic bacteria in various agents are 7 billion/g,0.128billion/g,24.7 billion/g.4.The research to PSB solid inoculants purify the farming water.The experiment com pares the effect among different Solid-type agents and carrier.It shows that after 48h of the purification,the activated carbon concussion agents could achieve the 44.07%removal of Ammonia and 81.89%of Nitrite nitrogen in the farming water.There is the order of different agents according to the purification effect it concluded:Oscillation agents group>Direct adsorption group>Carrier group.
