Whitmania pigra Whitman,which belongs to the arhynchobdellida Haeno pidae,vermiculite(Hirudinea),Annelida in classification of animal phyla,,all the dry leech was used as medicine,in the treatment of cardiovascular and cerebrovascular disases with significant effect,and it is also the main varieties in the Chinese leech cultivation.In recent ten years,in order to fill vacancies in the market,the development of leech cultivation is fast,has made remarkable achievements.Firstly,it had met the market demand in some degree,to ease the contradiction between supply and requirement.The leech cultivation had become a hot area and there are greatly small dozens of breeding base in Jianghuai area,the annual production could get one hundred tons,and it fills the lack of the market in reducing of wild resources.The second is the leech cultivation has a certain scale,the industry has been initially formed;Yangzhou has built 2000 mu breeding base,and its head company has achieved considerable success as the core component of the"Tongxinluo" drug development in the leech.The third is reducing the requirement of the wild leech resources,protect the wildlife resources;The place once is the traditional area which teemed with leech,because of overfishing,the leech population has declined,and the place now even cannot be found the leech.After the leech cultivation has greatly enlarged breeding area,the tension between supply and demand has been eased,and the phenomenon of overfishing was reduced,and the wild resources are protected.The last is the breeding skills has a certain foundation,the cultivation is becoming more and more mature;in the last ten years,the industry forward in twists and turns.With the leech market ups and downs,the industry has also been honed.Although the knowledge of leech is still less,we are lack of enough understand-ding,too.But leech cultivation achieved certain results as before,the skill of net cage culture is applied very universally,and the management is relatively reasonable.At the same time,in the fishery,large-scale indoor and intensive aquaculture has developed.The indoor is culture not only very mature application,but also improve the breeding density,survival rate,the utilization rate of resources and has a very good experience in water treatment,density,yield issues on,which gave us inspiration.This experiment mainly examine the growth of seedling and adult period from the survival rate and the weight two indicators,using the method of orthogonal test design,collection of water quality data,records the using of foods and drugs,observes the living habits of leech,all that is in order to analyze the influence factors on the growth of leech,find the key factors of interference.And it will find out the corresponding countermeasures,so that we can change the situation,of the inefficient field farming,to promote the development of the industry.This experiment is mainly divided into two stages,the first stage is the seedling stage,this stage is that cocoons were put and they adapted to the environment and they gradually grow;the second stage is a leech breeding stage,this stage is that seedling weighted gradually to adult leech.Factory seedling survival rate was 55.10%,the weight is 0.105g.Five factor experiment design----density,changing the water way,water depth,habitat,saponin.And after treatment,the orthogonal test results showed that the survival rate of the best group is 800000 breeding density,10cm water depth,all-changing water way,no habitat,put saponin,the influence rank from important to unimportant is as follows:the habitat,water exchange method,density,saponin,water depth.The best group of weight is:micro flow water,no habitat,800000 breeding density,20cm water depth,put saponin,the effect size is in turn:density,habitat,saponin,changing the water and the water depth.After the treatment of SPSS 17.0(including correlation detection,T test,ANOVA test and so on),the survival rate and the level of nitrite was closely related,and the weight and dissolved oxygen value is most closely;two independent samples t test,different water depth,density,saponins,habitat,changing the water have little influence on the water quality,it means that did not cause the different water quality.The survival rate of the indoor adult leech cultivation is 18.47%,the weight is 1.13g.Five factors of experiment design is changing the water way,depth,density,antidote,aeration,through the orthogonal experiment design and analysis,the survival rate of the best group is:all-changing water,no aeration,150000 breeding density,30cm water depth,antidote,the size of five factors on the experiment are in turn:density,changing the water,the depth of water,aeration,antidote.The best group of weight is:all-changing water,aeration,250000 breeding density,30cm water depth,no antidote,the effect size is in turn:aeration,water depth,water exchange,antidote,density.After the treatment of SPSS 17.0(including the correlation test,T test,ANOVA test and so on),survival rate and ammonia nitrogen is most closely related,but the body weight and the dissolved oxygen is most closely related;two independent samples t test that found the influence of five factors on different levels of water quality did not cause different water quality.The purpose of this study is in the orthogonal experimental design factors based on and found the relationship between the factors of Whitmania pigra growth influences the survival,water quality and Whitmania pigra Whitman to,aimed at through the research for leech large-scale factory farming reference,promote the development of leech farming.The significance of this study lies in the fact that experiments by orthogonal test design found for water,density and other factors of Whitmania pigra Whitman affect larger,and nitrite,the oxygen and other water quality indicators of the quality of the survival of Whitmania pigra growth also has a significant effect;it is a factory farmed Whitmania pigra water refers to the standard monitor and control and farming equipment design provides some guidance.
