In recent years,occurrence of Haemophilus parasuis disease,clinically characterized by fibrinous polyserositis and fibrinous arthritis,is common in swine production.Haemophilus parasuis has frequently been isolated from suffered animals in laboratory,and its pathogenesis is unclear.In this study,comparison proteomic and immune proteomic of two strains of Haemophilus parasuis serotype 4 isolated by our laboratory were analysed,expecting to explain the pathogenesis theoretically.1 Establishment and majorization of the 2-DE for the Haeumophilus parasuis2-DE reaction suitable for determination of Haemophilus parasuis was established and majorized by different protein extractions,different sample volums and isoelectric focusing(IEF).The Grinding-ultrasound-lysis method was used for extraction of proteins.The optimized parameter of protein lysis buffer were 7M Urea,2M Thiourea,4%CHAPS,40mM DTT and 2%IPG buffer,2D clean-up kit and DeStreak reagents were used,and the two-dimensional polyacrylamide gel electrophoresis was dyed with silver staining method.The optimized sample volume is 500?g for 24cm drystrip,IEF was determined at 30V 12h,100V 2h,200V 1h,500V 1h,1000V 3h,2000V 2h,6000V 3h,60000 Vh,500V 10h.2 The research of comparison proteomic of Haemophilus parasuis strains serotype 4With 2-DE and PDQest 8.0 software,different expressed proteins were seperated and analysed.The mass spectrum of these proteins were obtained by MALDI-TOF-TOF mass spectrometer,and 10 different expressed proteins were finally identified by protein search software Mascot2.3.02.3 The research of immune proteomic of Haemophilus parasuis strains serotype 4The total expressed proteins of stain A and stain B of Haemophilus parasuis serotype 4 were separated by 2-DE.With positive rabbit serum antibodies,immune response proteins were determined by Western-blotting.Compared to mass spectrum results,13 proteins were identified to be the same between strain A and strain B.And 18 proteins generated from strain A and 12 proteins generated from strain B were also identified separately.
