L-carnitine is a common nitrogen non-protein molecules in the animals which widely used in functional food additives,feed additives and pharmaceutical fields.1.The establishment and optimization of column L-carnitine derivatives-HPLC(High Performance Liquid Chromatography)detected carnitine.By investigated the effort on the mobile phase,column temperature to the chromatographic peak,Method of sample handling and derivatization reaction conditions were optimized,and the corresponding HPLC method was established:Kromasil Sil chromatographic column(250 mm x 4.6 mm,5 microns);Mobile phase:acetonitrile-citric acid buffer(90:10,V/V),0.25 mL per 1 L liquid phase joining triethylamine;Column temperature 35 ?;Detection wavelength of 260 nm;20 ?L sample quantity.After comparison of two different concentrations of derivative reagent,select 10%of bromobenzene acetyl bromide as derivative reagent;the L-carnitine in best bath conditions were determined as follows:carnitine 70 min at 70 ? water bath.Methodological investigation results show that,it is high specificity and good repeatability,the linearityand precision can be met the standard requirements,it can be analysis method for the L-carnitine.2.pharmacokinetics study of L-carnitine in tilapiaThis experiment compared the different dose pharmacokinetic regularity of carnitine in the tilapia and the same dose of carnitine pharmacokinetic in different specifications of tilapia.pharmacokinetic parameters were calculated by non-compartment method of WinNonlin 5.2 Practical Pharmacokinetic program software.The results show that,the drug dose of carnitine and test specification of tilapia fish all affect the pharmacokinetic parameters.To 12 mg-kg-1 and 120 mg-kg-1 dose mouth filling,12 mg-kg-1 and 120 mg-kg-1 body weight dose group of peak time are the same,the clearance and average residence time have no significant difference(?>0.05);but the dose of 12 mg-kg-1 body weight group of peak concentration(82.28 ?mo1-L-1)is significantly higher than 120 mg-kg-1 body weight group of peak concentration(21.27 umol/L),medication in the area under the curve(96.95 h-?mol-L-1),medication in the area under the curve(431.06 h-?m1-L-1).The results demonstrated that L-carnititine influenced by the dosage.The above two of the clearance and average residence time have no significant difference(?>0.05).This experiment compared the same dose of carnitine pharmacokinetic.in different specifications of tilapia.the peak concentration(Tmax)of carnitine in the tilapia?elimination half-life(ti1/2)and clearance(ClZ/F)along with the age increase becomed smaller,but the difference was not significant(p>0.05).As showed medication in the area under the curve,the absorption of drug distribution in the mature fish is more widely than the Immature fish.This paper for the first time measuremented the L-carnitine levels of plasma in the tilapia.Established the detection of carnitine in plasma by HPLC method,compares the carnitine pharmacokinetic regulation of different doses and different specifications of tilapia.This paper can provide important theoretical basis for analysis the content of these compounds in aquatic products and for the food nutrition and safety assessment,also provide the necessary premise and theory for the research and development of L-carnitine.
