The objective of the study was to extract and identify the pigment in Atrina pectinate and Atrina vexillum,we chosed the two kind of pen shell for experiment.The shells of Atrina pectinate and Atrina vexillum were dried after flesh removal,cleaned for shell,crushed,treated in HCl acid for hydrolyzation,heated in water bath and removed excess fat in Ether;Finally,blackparticles were extracted.The extracted black particles were dissolved in 0.01mol/L NaOH dissolution and ultraviolet absorption spectra were generated by an ultraviolet spectrophotometer.The maximum absorption peak was at 240 nm andthe absorption values decreased with the wave increasing within 190-500 nm,which sugested that the black pigment was melanin.Moreover,fourier transform infrared spectroscopy scanning revealed characteristic absorption peaks that 35003300 cm–1?16701600 cm–1 and 11001000 cm–1,which was consistent with infrared scanning features of eumelanin.Therefore,the extracted black particles from Atrina pectinate and Atrina vexillum shells were proved to be melanin for the first time.Melanin is not soluble in water,but only soluble melanin is useful in functional verification.Through acid-base method combined with ultrasonic crushing method,we successfully obtained the soluble melanin from the shells of Atrina pectinate and Atrina vexillum.We tested the black substance by ultraviolet spectrophotometer and fourier transform infrared spectroscopy scanning,and found that there was no different for the chemical property of melanin after ultrasonication.However,the solubility of melanin was improved.The experiment laid the foundation for the functional verification of the melanin of Atrina pectinate and Atrina vexillum.There are also shortcomings of this study,such as,the extracted melanin was still the crude products and further purification experiment was needed.In addition,the purity of the melanin can't be measured accurately,perhaps because the melanin itself is a mixture.
