Abstract/Summary:
MD is a common malignant lymphoma infection caused by MDV.To further study the mechanism of MDV tumorigenesis.In this study,animal models were constructed by infecting SPF chicken with MDV highly strain GX0101:(1)1 day old SPF chicken were divided into experimental group and control group of 40,Each chick group was injected intraperitoneally with 200 ?L of GX0101 strain.In the control group,each chick was injected with CEF primary cell fluid.The liver,spleen,lung,kidney,glandular stomach,thymus,brain,pancreas and other tissues and blood samples of the experimental group and the control group were taken on the 7th,14 th,21st,30 th,45th and 60 th day.The expression levels of meq and g B in the blood were measured by fluorescence quantitative PCR,and the histopathological changes were observed by HE staining.(2)The proteomics of the spleen of the experimental group and the control group after 45 days were analyzed by Label-free technique,and GO analysis and KEGG analysis.(3)The relative expression of PRNP was measured by fluorescence quantitative PCR in the experimental group and control group at different time points after injection of GX0101 and CEF cells.Through this study conclude that:(1)Fluorescence quantitative PCR showed that MDV marker gene meq and g B had the highest expression at 45 days after injection of GX0101,and multiple tissues were observed in 45 days.The expression of meq and g B was correlated with tumor formation.(2)Compared with the control group,690 proteins were upregulated and 390 downregulated.GO analysis note shows that these differential proteins are involved in biological processes that are mainly metabolic processes,transport and redox processes.KEGG analysis showed that these differential proteins were mainly related to cancer-related pathways,prion related pathways,signaling pathways and other pathways.(3)The expression level of PRNP gene was correlated with MDV infection,and the expression of PRNP gene was higher in the tissue of the experimental group than in the control group.The expression of PRNP gene in the tissue of the injection group changed with time,reached the peak at 45 days,then decreased to 60 d and was at the lower transcription level.
