Fangji Menispermaceae pLant tetrandrine(Stephania tetrandra S.Moore)is the dried root In the history of medicine.In China,onLy contained in the "Shen Nong's herbaL cLassic" grass,Listed as the goods.Moore was born in Mountain hiLLy Land,grass or forest edge,and generaL growth 5~10 years after excavation can dig,seLf recovery abiLity weak.Tetrandrine depends entireLy on the wiLd resources suppLy market in recent years due to a continued high price of tetrandrine,tetrandrine are disordered craziLy digging.This paper coLLected from 14 different areas of Fangji,to "Chinese Pharmacopoeia" as the standard,compares the differences between different areas of Lacking moisture,ash and extract content;compare the change of carbohydrate;and to the "Chinese Pharmacopoeia" as the basis,the contents of the main function components Fangji tetrandrine and fangchinoLine were determined;and the 14 origin Herbs Based on fingerprint by HPLC method,the above indexes,the differences in the quaLity evaLuation of Chinese medicinaL materia Ls with different effects,appLication and protection of Fangji provided theoreticaL basis.The contents and resuLts in this paper incLude the foLLowing aspects:(1)According to the 2015 edition of "Pharmacopoeia" method Chinese records,the content of Fangji in 14 different pLaces of moisture,ash and extract were determined,the difference anaLysis and dendrogram constructed using SPSS19.0 software.The resuLts showed that: different habitats in water on Menispermaceae meet Pharmacopoeia requirements shaLL not exceed 12%,in 14 areas in Anhui Mount Huangshan onLy cLose to the Limit;in totaL ash,14 herbs are in Line with the Pharmacopoeia provisions shaLL not exceed 4%,and the differences in the content of Less,14 production of Fangji pieces processing for aduLterated or contaminated;in the extract,content is significantLy different,with water,the individuaL origin of its Low water content,whiLe the extract content is high,shows that the two are Linked.(2)Using UV spectrophotometer to detect the different origin of Fangji totaL sugar and reducing sugar and poLysaccharide content,difference anaLysis and dendrogram constructed based on the SPSS19.01 software,the use of statisticaL methods to compare the variation of carbohydrate content.The resuLt shows the content of sugar in different areas of Fangji have obvious difference,the content of reducing sugar in the biggest difference,Zhejiang more than the potentiaL of reducing sugar content higher than the average content of about 2 times,and the Zhejiang Lanxi and Jiangxi Yongfeng is Lower than the average content of about 3 times.Hubei and Zhejiang,Changshan,the Largest difference in totaL sugar,Zhejiang,Changshan and Anhui,the Largest difference between the poLysaccharide.(3)The content of tetrandrine in different areas of tetrandrine and fangchinoLine by HPLC determination,SPSS19.01 software is used to anaLyze the differences and buiLd a dendrogram,The resuLts showed that content of Fangji 14 areas are consistent with the requirements of Chinese Pharmacopoeia(not Less than 1.6%),the contents of 3.43~5.08 in different areas;and Lacking content differences,anaLysis that may be the cLimate,geographicaL environment and the origin of pLanting;aLso coLLected reLevant medicinaL materiaLs saved,storage environment may change from the composition.Therefore,in order to stabiLity of Fangji appLication,estabLish pLanting system specification,and the standard way of Chinese herbaL medicine circuLation.(4)The physicaL and chemicaL indicators of Fangji indicators using SPSS19.01 software for correLation anaLysis and principaL components,and constructs the comprehensive cLustering of main chemicaL components in the dendrogram based on.The resuLts showed that moisture,ash,extract,reducing sugar and tetrandrine variation coefficients were above 25%,indicating the difference between different regions was Larger,the maximum is 39.14%,reducing sugar,indicating different origin between the reducing sugar content of the biggest difference;at the same time the correLation anaLysis showed that the 4 principaL components cumuLative variance contribution rate is 82.3383% PC1,the Largest contribution rate of PC1,mainLy incLuding water,tetrandrine and fangchinoLine and the totaL amount of basic,can be used as the main factors of Lacking of evaLuation.(5)By the method of HPLC in different pLaces were Lacking the fingerprint,and combined with the peak area of totaL components by cLuster anaLysis,to evaLuate the different origin of fangji.Using AgiLent EcLipse XDB-C18(250 mm * 4.6 mm,5 m)coLumn,acetonitriLe-0.1% formic acid soLution as mobiLe phase,fLow rate of 1.0mL-min-1,the coLumn temperature was 30 degrees,the detection waveLength of 282 nm,and combined with the software of spass 19 to totaL peak area by cLuster anaLysis.The resuLts showed that 14 sampLes of Fangji 9 common peaks.Among them,6 Letters for fangchinoLine,peak 7 for tetrandrine;10 origin fingerprint simiLarity was above 0.9,the same chemicaL composition,the germpLasm stabLe,not affected by the pLanting environment;a totaL of 9 peak area obtained by cLustering,cLustering resuLts wiLL not be the same areas of medicine are cLustered,indicating that the chemicaL constituents of different regions are different,but the difference is not the medicine compLeteLy independent,and can be more obvious that the chemicaL composition of Fangji is not affected by the environment,the difference did not reach the quaLity.
