Color is one of the important indicators of wine,anthocyanin as a coloring material affect the convergence and stability of wine,the study found that anthocyanins have many physiological activities and benefit to human health.The western region located at the golden latitude of the world grape cultivation areas,and has suitable climatic conditions,become China’s wine grape cultivation of high-quality producing areas,but the wine color is shallow,easy to aging and unstable.This experiment selected the Merlot grapes,sprayed MeJ over leaves at veraison and sampled at different times.Analyzed the effect of MeJ on physicochemical parameters of grape.Researched the changes of anthocyanin content in grape by HPLC-MS and the effect of MeJ on anthocyanin synthetase.Brewed wine after harvest,the effect of Me J on physicochemical indexes and anthocyanins of the corresponding wine was analyzed.Selected the best spray concentration,the aim was to provide theoretical basis for the application of exogenous MeJ to improve the content of anthocyanins.The main results are as followed:1.With the maturity of the grape,grain weight and soluble solids gradually increased,slightly decreased when matured.The total acid content gradually decreased and corresponding pH gradually increased,after treatment with different concentrations of MeJ showed a certain difference.The total anthocyanin content began to accumulate rapidly from the beginning of the veraison and continued to rise,but decreased slightly when matured.After MeJ treatment,the content of total anthocyanins increased with the increase of treatment concentration(P<0.05),the total anthocyanin was the highest after 10 mM treatment.2.A total of 20 anthocyanins were detected by HPLC-MS in grape peel,formed glycosides with glucose,which is the highest content of malvidin-3-O-glucoside and its derivatives.The total amount of anthocyanins was consistent with the trend measured by spectrophotometry,the total amount continued to rise and has a slight decline in ripening.The total amount of anthocyanins after 10 mM MeJ treatment was significantly higher than control,the trend and difference of Monomeric anthocyanin and acetylated derivatives were consistent with total anthocyanin content,however,the accumulation time of coumaroyl derivatives after treatment with MeJ was earlier.3’5’-substituted anthocyanins grew faster and the content was significantly increased,3’-substituted anthocyanins accumulate much more later,after MeJ treatment,there were significant differences in earlier stage,but the difference was not significant later.3.The key enzymes of anthocyanin synthesis PAL and UFGT showed significant difference in the short-term and long-term sampling results after MeJ treatment,and the long-term sampling activities showed a tendency to rise first and then decrease.CHI showed significant differences in the short-term sampling results only on the third day,and the long-term sampling results showed significant differences all the time.The difference of DFR in the short-term was not significant,followed by aging,activities increased,the difference was significant,to the late decline,the difference was not obvious.The correlation analysis showed that the total anthocyanin content was positively correlated with PAL and UFGT.4.There was no significant difference in the degree of alcohol,total acid and pH in the wine which brewed by MeJ treatment grape,the color related indexes showed significant difference.23 anthocyanins were detected in the wine,and 10 mM treatment showed significant difference on various anthocyanins,the anthocyanins of 3’5’-substituted increased significantly after treatment,while 3’-substituted anthocyanin increased less. |
