Chloroplasts of plant is an important organelle of photosynthesis and energy conversion,it's also an important carrier of female genetic information.As semiautonomous organelle,it is inherited in a maternal manner.Maternal inheritance has its own unique characteristics,good safety,and can avoid gene leakage caused by gene leakage.Compared with the nuclear genome,the cp genome is smaller in size and more convenient to clone and genetic manipulation.The chloroplast genome(cp)has multiple copies in each cell,which can ensure high expression of genes targeted to the chloroplast.It's said that chloroplast genome information is valuable in revealing the evolution of species,hybrids,evolution,and other aspects.With the rapid development study of other plant chloroplast genome,researches in chloroplast genome of herbage are also executed by leaps and bounds,but part of the herbage is limited to the chloroplast genome sequences available,not actually studied in-depth.Based on the importance of the research of chloroplast genome,in recent years,this paper collected the relevant information and summarized the latest research progress in this field from several aspects,isolation of chloroplast DNA,purification,sequencing and research progress of forage chloroplast genome to make everyone Learn more about chloroplast DNA purification and the research of status,future trends and prospects of forage chloroplast genome.In this paper,the sutdy assessed V.sativa cultivar Lanjian NO.3 and Medicago sativa,using DNase I purified chloroplast,sequencing by the second generation of high-throughput sequencing platform,and analyzed the chloroplast genome sequence of the two species.The results showed follows.1.The complete alfalfa chloroplast genome is a circular DNA molecule 128,574 bp in length,which has a special chloroplast structure containing only one copy of the IR region and GenBank accession number is KU321683.The alfalfa chloroplast genome encodes 109 genes,including 4rRNA genes,30 tRNA genes,and 75 protein-coding genes.2.V.sativa cultivar Lanjian NO.3 cp genome is 121,883 bp in length with only one copy of the IR region.It encodes 109 genes,including 4 ribosomal RNA(rRNA)genes,29 transfer RNA(tRNA)genes,75 protein-coding genes and 1 pseudogenes.V.sativa cultivar Lanjian NO.3 cp genome sequence has been registered in GenBank,and accession number is KU053796.3.Compared with other legume species,their chloroplast genes have few differences with the size,order and structure.The successful sequencing of V.sativa and alfalfa cp genome provides a solid foundation for chloroplast molecular biology and effectively promote genetic breeding and molecular evolution of the common vetch and alfalfa.
