Plants defense pathogens through multiple mechanisms.Systemic acquired resistance(SAR)is the most important part of disease resistance pathway.When physical and chemical barriers are broken by pathogens with type III secretion system,they trigger effector-triggered immune(ETI).ETI often associates with the biosynthesis of salicylic acid(SA)locally and systemically,and induces systemic acquired resistance(SAR).If plants exposed to repeat pathogenic infections,genes could alter response to improving resistance to future exposures like ‘memory'.This research explored the function of ADH of disease resistance in Arabidopsis.Phenotype assays,bacterial colony count assays,ADH activity assays and RT-qPCR assays were performed in WT and adh1 mutants treated with Pseudomonas syringae DC3000,SA and cold conditions.(1)ADH associated with disease resistance.During DC3000 treatment,adh1 resulted in an intense hypersensitive response than WT.ADH activity assays and RT-qPCR analysis showed that DC3000 induced the expression of ADH.It revealed that ADH associated with disease resistance.(2)ADH was induced by SA,and associated with SAR.ADH activity assays and RT-qPCR analysis showed that SA induced the expression of ADH1 up-regulation.It implied that ADH involved in SAR pathway.(3)ADH affected the expression of NPR1,the central element of SAR.RT-qPCR analysis showed a difference of SAR relative genes,NPR1,PR1 and WRKY53 under SA treatment between WT and adh1 mutants,especially the expression of NPR1.(4)ADH associated with disease resistance memory.During repeat DC3000 treatments,ADH showed different expression patterns which indicating the phenotype of ‘memory'.ADH exhibited a higher expression in second DC3000 treatment than it in first time.It implied that ADH was a memory gene of disease resistance.(5)ADH affected disease resistance after 4? cold treatment.After 4? cold treatment,a new DC3000 infection made plants produced a feeblish hypersensitive response.The expression of ADH and SAR related genes of disease resistance were inhibited by cold.Above results showed that ADH associated with disease resistance in Arabidopsis,it affected the expression of SAR related genes,and ADH1 was a memory gene of disease resistance.This study provided a theoretical basis for elucidating the function of ADH in plant disease resistance,and also provides some new insights for plant disease resistance research.
