In order to understand the popular and distribution of canine source to E.coli of PMQR genes in Shihezi,Xinjiang.Sterile collection the rectal swab of diarrhea and healthy pet dogs from three animal hospitals,by experimentation of bacterial isolation and identification,biochemical identification,16 Sr DNA amplification,and drug resistance detection of 14 kinds of common antibiotics.At the same time,using the PCR technique determination of the Escherichia coli to Phylogenetic Group,and screening canine source to Escherichia coli of PMQR gene.Research results are as follows:1.63 strains Escherichia coli have been isolated and identify,and 16 SrDNA amplification from 75 samples rectal swab of diarrhea pet dogs and healthy pet dogs from three hospitals in Shihezi,Xinjiang.There are 45 strains came from diarrhea dogs and 18 strains came from healthy dogs.The total separation rate was 84 %.The hemolytic and pathogenic experiment showed that most of β-hemolytic strains were came from diarrhea dogs.After intraperitoneal inoculation the mice with pure cultivate of E.coli showed that β-hemolytic could lead mice in less than 50 h all died.2.Detecting the 63 strains Escherichia coli with Phylogenesis Group using the multiple PCR,most of isolates were distributed in group A(28 strains,44.44 %),followed by group B2(22 strains,34.92 %)and D(10 strains,15.87 %),a very few of isolates were distributed in group B1(3 strains,4.76 %).Almost all distributed in group B2 and group D of canine source to Escherichia coli was have pathogenic.3.Choice of 14 kinds of common antibiotic resistance testing.Results showed that,the highest drug resistance rate to tetracycline by 85.71 %(54/63),followed by lincomycin and enrofloxacin,the drug resistance rates were 73.02 %(46/63)and 68.25%(43/63).Strains to florfenicol is most sensitive,the drug resistance rates only 4.76 %(3/63).Other antibiotics resistance rate distribution in 17.46 %~ 65.10 %.80.95 %(51/63)isolates resistant to four kind of above and four antibiotics.Most isolates were resistant to 9 kinds of antibiotics have resistant,accounting for 44.44 %(28/63).Followed to 6 kinds of antibiotics resistance,accounting for 36.51 %(23/63).3.17 %(2/63)isolates had the resistant to 14 kinds of antibiotics and 4.76 %(3/63)isolates were sensitive to 14 kinds of antibiotics.4.Detecting of 7 kinds of quinolone resistance genes by PCR,the detection rate of qnr gene was 36.51 %(23/63).The qnr A gene had the highest detection rate,accounted for 20.63 %(13/63).Followed by qnr S genes,the detection rate was 11.11 %(7/63).The detection rates of qnr C and qnr D genes were 3.17 %(2/63)and 1.59 %(1/63).Detecting rate of aac(6’)-Ib gene was 12.70 %(8/63).The qnr B genes and qeq A gene were not detected.There were 11(17.46 %)isolates contains to two kinds to PMQR gene;only one isolates contains to 3 kinds PMQR gene,the detection rate was 1.59 %.This study for the first time to conduct testing to the PMQR gene of Escherichia coli from canine source in Shihezi,Xinjiang.And extended the canine source to E.coli of the epidemiological data about PMQR gene.Providing data to support for the study about drug resistance from canine source of E.coli.Then offer scientific basis for the correct selection quinolones in the prevention and treatment of canine bacterial diseases at Veterinary Clinic.
