Object: Comparing the effects of barcode on the identification of Salvia miltiorrhiza,so as to provide the molecular basis for the identification of Salvia miltiorrhiza.The expression of key enzyme gene in the synthesis of Salvia miltiorrhiza liposoluble composition was studied in order to find out the most important factors.And then make a fundamental contribution to the biosynthesis of the liposoluble components of Salvia miltiorrhiza and to solve the drug source problem of Salvia miltiorrhiza.Methods: The UV was used to test the content of total flavonoids and total phenolic acid,the antioxidant capacity.The contents of main effective components were determined by HPLC.Then,from the perspective of bar-code,we selected ITS2,matK,rbcL and psbA-trnH to study the research materials,and quantitative PCR were used to study the key enzyme genes.Finally,the obtained data were analyzed systematically.Main results:(1)Through the comparison of the appearance,the content of total flavonoids,total phenolic acid and antioxidant activity of these 10 Salvia miltiorrhizas,we found that there are some differences in appearance and intrinsic chemical composition of the studied materials.Lay the material foundation.(2)In this study,psbA-trnH and ITS2 were spliced to identify 10 Salvia miltiorrhizas.Then we found that 10 different Salvia miltiorrhizas could be distinguished,which provided a molecular basis for demonstrating the differences in the experimental materials of this study and provided a new identification method for Salvia miltiorrhiza.(3)Through the correlation study on the content of tanshinone components with the expression of key enzyme genes,we found that the correlation between DXS1,DXS2,GGPPS,CPS,HMGR with tanshinones content is high,while the correlation between HDR,KSL with tanshinones content is low.Conclusion: The combination of psbA-trnH and ITS2 provides a new method for the identification of Salvia miltiorrhiza.DXS1,DXS2,GGPPS and HMGR,MCS,CPS may be important intrinsic factors affecting the content of liposoluble components of Salvia miltiorrhiza,while HDR,KSL show little effect on it,which provide a reference for improving the content of liposoluble components of Salvia miltiorrhiza from the molecular angle.
