Agaricus bisporus is the widest human cultivated straw-decay fungus in the world.Additionally,our nation is the biggest export country in Agaricus bisporus in the world.Casing layer is an important part for the cultivation of Agaricus biaporus.Casing layer is not only the main place for Agaricus bisporus growth and development,but also it is the essential material basis.In this paper,Agaricus bisporus was used as research object to study the effect of P.putida on the growth of Agaricus bisporus.With fluorescence labeling technique,it explores the effect of P.ulida on its colonization ability,yield and quality.Considering all the factors,it makes sure that what appropriate the microbial agent can promote the growth and development of fruiting bodies of Agaricus bisporus.Specific results are as follows:1 The attribution of P.putida TK3The logarithmic phase of P.putida TK3 is ranged from 8h to 14h,the bacteria in this period are most sensitive to antibiotics.P.putida TK3 was resistant to Ampicillin,but it does not possess Kana resistance.It was resistant to Gm,for low concentration(10 ppm,30 ppm,50 ppm),but for high concentration(80 ppm,100 ppm)Gm it does not possess resistance.After the positive identification,the pBBR1MCS5-RFP was successfully transformed into the cell and it did not affect its attribution.2 Co-cultivation experiment of P.putida TK3 and Agaricus bisporusThe paper found that P.putida TK3 and P.putida TK3(rfp)respectively co-cultivation with Agaricus bisporus could promote the growth of mycelia of Agaricus bisporus by measuring mycelium diameter(co-culture plates),mycelium length(in vitro co-culture),mycelial biomass(liquid co-culture).Red fluorescent protein did not affect the growth ability of P.utida TK3.Agaricus bisporus 192 varieties grew better in the co-cultivation method compared with Agaricusbisporus 2000 varieties.3 Effect of P.putida TK3(rfp)on Agaricus bisporus growth,yield and qualityBacteria accounted for the most proportion among microorganisms in casing layer.With the increasing concentration of microorganism inoculation,the number of bacteria was also increasing.However,the number of Agaricus bisporus primordia is slightly decreased in its formation period.The number of P.putida TK3(rfp)stabilized in the casing layer after successful colonization.Adding certain concentration of P.putida TK3(rfp)into casing layer could increase mushroom production.When spraying EM mixing with P.putida TK3(rfp)than only adding EM,the yield is higher than before.The concentration of a particular P.putida TK3(rfp)could improve the quality of Agaricus bisporus.Taking the soil microbial community,yield,quality into a commercial account,it is strongly suggested that 30mL TK3(rfp)agent(the number of cells per grams of casing layer was 106)was used as a new measure after covering for high yield and quality of Agaricus bisporus. |
