The rapid development of plant tissue culture,so how to carry out automation to improve seedling quality,growth rate and reduce labor intensity finally reduce the cost of production has become one of the key technology of plant tissue culture.Therefore,the intermittent immersion bioreactor system(Temporary Immersion Bioreactor system,TIBs)came into being,it is one of the earliest systems used in plant tissue culture and production of secondary metabolites of cultured material in liquid medium of intermittent immersion culture,in the past 30 years,researchers have to design more perfect,the training material is placed in liquid medium rest for training,so that it can be used for mass production in order to achieve the purpose of rapid propagation,this culture plantlets have stronger resistance,can improve the survival rate of the late seedling domestication stage,eventually reduce labor costs.The research of lifting liquid immersion intermittent using self-developed training device,banana,sugarcane,cassava,three kinds of tissue culture seedlings as test materials,the research on the immersion frequency,sucrose content,inoculum density and inoculation effect of materials following algebraic parameters such as rate and height on proliferation of plantlets,the rooting after transplanting seedling cultivation.The results are as follows:1.The herbs banana,sugar cane and bush plant cassava for static liquid solid,intermittent immersion culture,to study the three kinds of training methods,the results were as follows:the most suitable culture method of plantlets of banana and sugarcane were liquid intermittent culture,proliferation multiples of TIBs system for banana were 7.53,were 2.43 times of the solid culture proliferation,in rooting culture,solid culture root number was 5.80,slightly better than the TIBs culture 5.20.The multiplication time of TIBs system was 11.5,which was about 3.03 times that of solid culture.The number of TIBs rooting was 4 in rooting culture,which was better than 3.40 in solid culture.The most suitable cultivation method for subculture of cassava was solid culture.The multiplication number was 3.40,the rooting number was 4.60,in TIBs,the multiplication number was 3,and the rooting number was 3.20.2.In the TIBs system of different intermittent frequency on different seedling test,the results of the study were:the highest cultivate proliferation ratio of the banana TIBs intermittent frequency was 15min/2h,proliferation ratio was 6.42,the highest plant height and stem diameter in intermittent frequency was 10min/8h.Optimum sugar cane TIBs system to cultivate the intermittent frequency was 10 min/8 h,6-BA the optimal concentration was 3 mg/L,proliferation ratio was 11.53;The optimum frequency of cassava intermittent culture was 10 min/8 h,proliferation ratio was 3.00.3.In the TIBs system with different effects of sugar on proliferation of different plantlets,the optimum sucrose concentration in TIBs banana,sugarcane,cassava plantlet proliferation are 30g/L,the sucrose concentration of 30g/L when the banana TIBs system to develop the highest proliferation rate is 8.62,the most suitable hormone concentration is 3mg/L6-BA+0.3mg/LNAA,proliferation in multiples of 6.73;the concentration of sucrose was 30g/L under the condition of sugarcane TIBs culture system optimal hormone concentration is 3mg/L6-BA+0.1mg/LNAA,the proliferation rate is 7.54;when the sucrose concentration was 30g/L and the hormone free,the proliferation rate of cassava was 3.00.4.In the same conditions,TIBs system is most suitable for banana and sugarcane tissue culture seedlings,the density of culture is 10 strains/bottle,and the multiplication times are 7.30 and 8.20 respectively.5.The banana plantlets from fifth generation to ninth as the tested materials were cultured,the most suitable Banana Plantlets in the TIBs generation system relay proliferation subculture materials for the seventh generation,the proliferation rate is 7.17;the sugarcane seedling from fourth generation to seventh as test material,TIBs material system is the most suitable for subculture proliferation sugarcane is the fifth generation,the proliferation rate is 7.32.6.Cultivation of different culture methods transplanting plantlets were cultured in TIBs system test,banana,sugarcane seedling transplanting survival rate is slightly better than that of solid and liquid culture,TIBs culture system of banana and sugarcane seedling transplanting survival rate were 90.63%and 82.22%,the survival rate was 81.82%,77.78%solid culture;culture is better than TIBs survival the rate of traditional cassava plantlets transplanting,TIBs culture transplanting survival rate was 0,the traditional seedling transplanting survival rate was 63.33%.
