Mongolian horse is one of the oldest species in the world.Has a good reproductive capacity and unique genetic.By measuring the testis can effectively predict the breeding ability of the male horse.Testicular development is an important physiological process of male mammals,producing sperm and secreting male hormones is the basic function of the testis.In order to understand the regulation mechanism of testis development and spermatogenesis of Mongolian horse before and after sexual from the genetic level,a new generation of high-throughput sequencing technique was used to establish the transcriptome library of testis tissue in Mongolian horse sexual maturity before and after the two stages of development.The results were analyzed by variable shear analysis and differentially expressed genes.GO gene enrichment analysis,KEGG Pathway analysis and transcription factor analysis were performed to find testicular development and sperm production related GO function enrichment,KEGG pathway and related gene.Finally,Application of Real-time quantitative PCR method to detected the mRNA expression levels of TSSK2,SPATA6,CDC20 and MAD2L1 genes in Mongolian testis were measured before and after sexual maturation.This study is of great significance for the selection of good use of male horse.Revealing the molecular mechanism of testicular development and sperm production in male equine,and obtains the following results:(1)24878 differentially expressed genes were obtained by transgenic gene expression in Mongolian mice before maturation,including 16237 genes and 8641 genes.These differential genes were annotated to 10019 GO function entries.Mainly in the spermatogenesis,male gamete generation,spermatid development and other biological processes closely related to testicular development and sperm production.(2)The differential expression of the gene was analyzed by KEGG pathway with testicular development and spermatogenesis,including Oocyte meiosis,progesterone-mediated oocyte maturation.(3)The expression of TSSK2 gene,SPATA6 gene,CDC20 gene and MAD2L1 gene were verified by real-time fluorescence quantitative PCR.The results showed that the expression of these four genes in the testis was more than 2 times higher than that in the testis before maturation;the difference is significant.The accuracy of the transcriptome sequencing results was demonstrated.
