Sequence Comparison Of Virulence Related Genes Of Parental And Its Attenuated Virus Strains Of Orf Virus

This study will Qingdao a diseased caprine field separation and preservation of ORFV/QD/2015 strains of fibroblast cells for 90 generations in the goat skin,according to the published Genebank gene sequence of ORF was designed and synthesized 4 pairs of specific primers and amplified the virulent ORFV/QD/2015 and ORF020 strains after 90 generations,ORF112,ORF117 and ORF127 genes fragment and cloned into PMD-19T vector and transformed into DH5 alpha competent cells.The recombinant plasmid was identified by sequencing,using DNA software Star sequencing results,biological information analysis.The results showed that orf virus could be cultured on goat skin fibroblast and produce cytopathic effect.Results the ORFV/QD/2015 plasmid sequencing of virulent and passage 90 virus gene sequence of 9 groups of ORFV and Genebank has been published in ORF020,ORF112,ORF117,ORF127 gene results ORFV/QD/2015 virulent and released strain virulence related gene homology,passage 90 strains and released strain virulence related genes the homology reduced,related gene of virulent and passage 90 strains of 4 groups of virulence nucleotide alignments showed that ORF020 gene had 7 amino acid mutation;ORF112 gene has 52 amino acid mutation;ORF117 gene mutation at amino acid 14;ORF127 gene 15 amino acid mutation.After comparison,it was found that the most variable nucleotide and amino acids were ORF112 gene,and the least was ORF020 gene.This indicated that the attenuated orf virus might be related to the variation of these virulence related genes.The antigenic indices between the two strains also varied significantly.These variations provide the experimental basis for differences between virulence related genes in virulence.
