| ObjectiveEnrofloxacin is a third-generation synthetic antibiotic fluoroquinolones,also a kind of exclusive antibiotic drug for animals.It has broad-spectrum antimicrobial effect and strong antibacterial effect.Therefore,it’s widely used in aquaculture of livestock and fish farming.However,enrofloxacin overusing can cause residual problem in animal foods,which harms to human health.Besides,trace enrofloxacin has ecological toxic.Presently,at home and abroad,the common methods of enrofloxacin detection in animal food include high performance liquid chromatography,enzyme-linked immunosorbent assay,capillary electrophoresis,and so on.These detection methods are costly,complicated,the methods have complex pre-treatment,also the application of environmental conditions were limited.Therefore,it is necessary to establish a rapid and simple method for the determination of enrofloxacin.In our study,CdS nanoparticles modified wire-coated ENR membrance selective electrode has been prepared,the potential analysis method has been established for the determination of drug content.MethodsTwo different complexes were prepared respectively by Bi I6-3 and sodium tetraphenylborate with enrofloxacin as two different electroactive substance.The two electroactive substance were made into electrodes with different materials,the enrofloxacin selective electrode with Nernst response and good performance was selected.In this case,CdS nanoparticles were used to prepare nanoparticles modified enrofloxacin electrode for the sake of improving the performance of normal enrofloxacin electrode.The electrochemical method was established to determine the performance of the electrode,finally,our electrode was applied for enrofloxacin detection in pannage and pig muscle.ResultsThe Nernst response of the normal wire-coated ion-selective electrode ranges from 1×10-1 to 1×10-5 mol/L,with a slope of 26 m V/p C and a limit of determination of 8.9×10-6 mo/L.The normal wire-coated membrane electrode was used to test the pannage samples,the RSD of the electrode was less than 3.7%.CdS nanoparticles modified wire-coated ENR membrance selective electrode has Nernst response that range from 1.0×10-2 to 1.0×10-6 mol/L,with a slope of 45 m V/p C and a limit of determination of 7.4×10-7 mol/L.Also,its operating life could reach 12 weeks,two more weeks longer than general electrode,the response time was shorter than 10 s when the concentration was ranging from 1.0×10-1 to 1.0×10-4 mol/L,while the response time was faster than 35 s under the concentration from 1.0×10-5 to 1.0×10-7 mol/L.The CdS wire-coated electrode was applied to the determination of ENR in pork samples,the results had demonstrated that our method has no significant difference contrasting with the standard way.ConclusionsIn our study,two kinds of normal wire-coated ion-selective electrode and CdS modified wire-coated membrance electrode has been prepared.CdS wire-coated membrance electrode was proved that it has a wider Nernst response,higher slope,lower limit of determination,shorter response time and more expanding p H application range comparing with the normal electrode.Also,our study provides a reference for optimization ideas of electrode and rapid detection of ENR in practical simples. |
