Rice is one of the world’s three most important crops.In Recent years,drought and soil salinity have been more and more aggravated,and they have seriously affected the rice production all over the world.The need for researching in drought and salt stress becomes more and more urgent.In this study,we found the two rice mutants 245em and 340 in Zhonghua 11 background were sensitive to drought and salt stress,and analyzed the reasons leading to this.For 245em,we found the spaces between two pavement cells were larger than wild type,and that the stomata density was increased,and that the stomatal closure extent had decreased in drought condition.These might fasten the transpiration speed of 245em.Moreover,the root system of 245em was weaker than that of wild type,and this could impair the water absorption of 245em.For 340,the morphology of stomata in leaf was abnormal,and the stomatal opening extent was increased.These would promote the transpiration.In addition,for detached leaves in 340,the water loss rate was faster,and the relative water content was lower than that in wild type.These indicate that the water retention ability of 340 has been cut down.In drought condition,that the rate of erected prickle hairs has decreased would promote the transpiration.Similar to 245em,the root system of 340 was also weaker than that of wild type.In our study,we also observed other phenotypes of 245em and 340.Both the mutants were dwarf,and the majority of agronomy features in 245em were decreased.For 340,the auricles were disappeared,and the angle between a leaf and the stem was larger.The angles between roots and the axis of root system were also larger.In Arabidopsis,PIPE can interact with DELLA protein.And OsPIPE,a homolog of PIPE in rice,may work in a similar way.In this study,I constructed a vector of proOsPIPE::GUS,and then got some transgenic rice plants.I found OsPIPE can express in the edge of leaves and especially in the veins of leaves.In addition,I found appropriate concentration of VB1,enhanced gas permeation,higher temperature in the regeneration,and dried transgenic calli before screening and regeneration can improve the speed and efficiency of regeneration.
